Objective : Study of Urumqi No. 10 cold spring in the bacterial community in the structure and function changes pre- and post- earthquake, the relationship and the study of its dynamic changes and 10 spring water and chemical factors. In order to find the response relationship between The bacterial community in the No. 10 cold spring in Urumqi and and earthquake,the structure and the function of the bacterial community before and after the earthquake in the No. 10 cold spring in Urumqi is studied,so did its relationship with the physicochemical factor of the spring. Method:Traditional culture method and BIOLOG micro tablet technology were applied to monitor the activity and diversity of function of the bacterial community before and after the earthquake in the level of cell,while the T-RFLP(terminal restriction fragment length polymorphism)method were used to analysis in the level of molecular.At the same time, PCA(principal component analysis) were adopted to analyze the correlation between bacterial community and hydro-physico-chemical factors of the spring water,while real-time fluorescent quantitative technique were employed to detect the density of thebacterial population of No. 10 cold spring in Urumqi.Result:Statistics showed that the culturable bacterial density was low.When the level of earthquake is under 2.8, the number of bacteria which could be cultured were more after the earthquake than before.When the level is higher than 3.0,the AWCD of the bacteria using carbon source and reacting to the chemical material is higher after the earthquake than before,so did the diversity of the bacterial community in No.10 spring in Urumqi. As it showed,during the testing period the hydro-physico-chemical indicators of No. 10 cold spring in Urumqi were fluctuating. Physico-chemical factors,which F-、HS、Hg、He、N2 were associated with N-Acetyl-D-Galactosamine 、 L-Serine 、 Inosine 、 N-Acetyl-D-Galactosamine 、Bromo-Succinic Acid significant positive( negative) correlation. Found in the chemosensitivity test of chemical sensitive material and environmental factors, there is a negative correlation between them, lincomycin and F ion was negatively correlated. And before and after the earthquake in Urumqi 10 spring, bacterial community diversity index in the sulfide(HS) and Simpson(D), Shannon index(H) showed a significant negative correlation with the index, CH4 evenness index(E) were significantly correlated, the correlation was not significant.The standard curve parameter in line with the basic parameter requirements, which R2 = 0. 993, S =-3.415, E=1.963. Without earthquake period the number of 16 S rDNA fragment copy of the bacteria in Urumqi No.10 spring is the highest, the post- earthquake’s is second, and the pre earthquake water bacteria 16 S rDNA fragment of the copy number is the lowest. Conclusion:The bacterial community of No.10 spring in Urumqi Changed obviously before and after the earthquake, and the results of the three methods trend present consistency. The use of carbon reacts with chemically sensitive substances, diversity index were associated with the 10 th springARTS factor has a certain relevance.Real time fluorescence quantitative PCR detection system has been initially established parameters consistent with requirements, and to be sensitive, rapid quantitative unknown template Urumqi No. 10 spring bacterial number.
