The initiation of embryogenesis, zygotic activation plays a substantial role in the whole embryo development。Understanding the molecular mechanism of zygotic activation makes great sense in improving the zygote development and subsequently the production of crops. This study has screen the genes regulating zygotic activation by mutant screening and construct the mapping population to clone the genes. The mainly results are as follows:1. Mutant genetics has been used to screen the embryo-lethal mutant after fertilization in the model plant Arabidopsis thaliana. Based on the previous work, we have got about 50 candidate genes related in zygotic activation2. One gene(FAC11) involves in zygotic activation has been rough mapped by positional cloning and another gene(FAC14) has been fine mapped, also two genes have been cloned(FAC2 and FAC17), subsequently one gene(FAC6) has been functionally elucidated in details.3. Two mutants(FAC2 and FAC14) have been finished the cytological analysis by DIC after HCG cleared, calculating the different ratio of embryo-lethal in each mutant. The results show that FAC2 and FAC14 are all the genes involve in early embryogenesis whose mutation can lead to the early lethal during embryo development.4. In the end, bioinformatics has been used to predict the structure and function of the protein encoded by the genes we have cloned and finished the construct of yeast two hybrid of FAC6 to screen the interactive protein.In this work, we have set a good basement for the other FAC gene cloning and functional analysis, meanwhile our work is an initiation for further molecular and genetic research of zygotic activation.
