Liliaceae belong to monocotyledon in angiosperm and widely distribute in the world. Liliaceae hold a lot of medicinal plants. It is important to use correct species for clinic usage, but it is not easy to identify them with traditional way accurately or quickly. This study chose 25 species as research materials from 18 genus which included 36 samples. We adopted the technique of DNA barcoding to identify the species. The experiment selected 6 candidated sequences and amplified them in all samples including rpoC1,rpoB,rbcL,matK,psbA-trnHfrom chloroplast and ITS2 from nucleus.DNA was successfully extracted from 34 samples and we got PCR products of 6 candidated barcodes. The result showed that different amplified efficiency was found in 6 candidated sequences: the rate of psbA-trnH, matK and rpoC1 could arrive to 100% while other sequences were 97%,85% and 79% respectively. In addition, the results of sequencing indicated that the rate of production in rpoB and ITS2 was greatly lower than other sequences.We obtained sequences from these samples and analyzed them with the soft of MEGA. Phylogeny trees were constructed with psbA-trnH, rbcL, matK and rpoC1 respectively. Results showed that single candidate sequence could not distinguish all the selected species while the combination of two or more candidate sequences could identify all the samples completely. The recommended combinations were psbA-trnH+matK,psbA-trnH+ rbcL and rbcL+ rpoC1.
