This text take Spirulina innermongoliansis for materials researched characteristics of the Sediment and Float of Spirulina cell, through studying environmental factors impacting the Sediment and Float of Spirulina cell, relation between the sediment and float of cell and transformation of main inclusion, and morphology change of gas vesicle. The results showed that the sediment and float of cell was mainly regulated by light with cell sediment under strong light and cell float under weak light. Temperature impelled cell to sink under strong light. Under strong light for a short time, carbohydrate accumulation in cell and gas vesicle atrophia resulted in cell sediment that was observed in 2-3 hours. Under strong light for a long time, with accumulation of massive carbohydrate and inflatable gas vesicle, the primacy reason for cell sinking was carbohydrate accumulation which was confirmed by test that cell would float with inflatable gas vesicle because of decrease of carbohydrate content which was a result of carbohydrate consumption by respiration. This showed that ecological characteristic of the sediment and float of cell was regulated by light, however, with lengthening of illumination time, cell sediment mechanism was adjusted which was a result of long-term adaptation to environment. This study yet showed that both accumulation and degradation of protein and adipose could not impel cell to float or to sink. Change of carbon source and nitrogen source during Spirulina innermongoliansis mixed culture little influenced the sediment and float of cell, however, carbohydrate accumulation because of nitrogen deficiency in culture medium resulted in cell sinking, which also proved that the sediment and float of cell significantly correlated with metabolism of cell carbohydrate. This paper comparatively researched extraction methods of carbohydrate in Spirulina cell, determination method of protein content and fouling resistance of mixed culture. The results showed that carbohydrate was more sufficiently extracted by hydrochloride extraction method; protein content was accurately determinated by Kjeldahl nitrogen method; mixed culture active with oxygen machine treatment by 5 min/3 times/d completely meets anti-pollution requirements.
