Arthrospira (Spirulina) innermongoliansis powder was taken as raw material , the conditions of hyrolysis by different enzymes and the Molecular Weight of hydrolysate were studied in this paper. The condition of refining the hydrolysate was also studied. Four enzymes (Alkaline-protease, Pepsin, Trypsin, Papain) were chose for hydrolysis. Based on single-factor tests analysis and response surface method (RSM), the conditions of each enzymatic reaction were determined and optimized. Variance analysis showed that the capacity of four proteinases for spirulina in their respective optimum conditions followed by Papain> Alkaline-protease > Pepsin> Trypsin, and the DH of Papain is significantly higher than the other three, the worst is trypsin.Compared the DH of four proteinases at the best condition, the DH of Papain is the highest one. Through the test of response surface with statistical analysis system, the optimum conditions were: temperature 62℃, pH6.5, enzyme concentration 4.5%, and time 3h.Under these conditions, DH was 36.18%.The colour of the hydrolysate is dark and the salt is too much. The activated carbon will remove the dark colour, based on the OD of hydrolysate and the reclamation rate of peptide, the optimum conditions were: activated carbon concentration3%, pH2.0, temperature 60℃, time 40min, the reclamation rate of peptide was exceeded 80%. Salt could be removed by ion exchange resin under the condition of the flow rate of 6vol/h, the hydrolysate reclamation rate and the desalinization are exceeded 80%.The distribution of Molecular Weight of hydrolysate was determined by Sephadex G-15. The results show that, in the products enzymolysed by Papain, peptides with molecular weight between 150-700u were 71.3%, below 150u were 24.7%, polypeptides were about 4%.
