Selenium is one of important elements to life, selenium is distributed widely but extremely unevenly on the earth, 2/3 of area in the world lacks selenium. Above 10 kinds of diseases and cancers are related to the lack of selenium. This situation seriously threatens the health of human beings and the development of animal husbandry. Plant is the key link in the ecological selenium and the inorganic selenium cycle and it is also the most important direct selenium resource for the human and the poultry.Using Arabidopsis thaliana as plant materials, the effects and mechanism of selenite on plant growth were investigated by a serial of experiment such as the establishments of Arabidopsis suspension cell line. the effects of selenite on growth of Arabidopsis suspension cell and seedlings of wildtype Col-4,cry1 and cry2 mutants. The main results were listed as follows:(1) The media for Arabidopsis thaliana suspension culture had liquid and solid two types. The typical S-like growth curve in a subculture cycle was obtained. In normal condition, a logarithmic phase of growth of suspension biomass occurred in 2-4 days after subculture. It was advisable to choose 10 mL/50 mL as diluting ratio for holding subculture cycle. Different concentration of hormones and sucrose in media greatly affected growth of suspension cells. 30g/L sucrose, MS with NAA0.5mg/L and KT0.05mg/L or B5 with NAA0.25mg/L and KT0.05mg/L were advisable to get the best status of Arabidopsis thaliana suspension cell growth.(2) Selenium nanoparticles have been synthesized in an aqueous solution by using chitosan (CTS) as a soft template, the average particle size of selenium is between 40~100nm. By co-culture with Arabidopsis thaliana suspension cell, the results indicated that organic selenium was less toxic and more easier to absorb than the inorganic selenium. When treated with 4mg/L of Na2SeO3 and 6mg/L of Selenium nanoparticles, the best growth status of the Arabidopsis thaliana suspension cell was obtained. It also contained the biggest amount of chloroplast.(3) The proper application of selenite to Arabidopsis col-4,cry1,cry2 could promote their growth. The seed germination was suppressed when treated with excessive 15mg/mL of selenite. while it promoted the growth at 1-2 mg/mL. The content of selenite raised with the increase of treatment concentration, when concentration of Na2SeO3 treatment is below 8 mg/mL. The activity of GSH-Px and SOD showed the same transformation rules. Treated with the same high concentration of Na2SeO3, Arabidopsis cry1 grew better than the col-4 and cry2, which indicated that high selenite-tolerance ability of Arabidopsis cry1 was higher than that of col-4 and cry2.
