Soil salinity is one of the major stress factors limiting the growth and productivity of crop plants in agricultural fields. Salt stress consisting of osmotic and ionic stress,and then the secondary stress such as oxidative stress arising by them, can disrupt homeostasis in water potential and ion distribution. This disorder of homeostasis occurs at both the cellular and the whole plant levels. Drastic changes in ion and water homeostasis lead to molecular damage, growth inhibition and even death. Plant salt-tolerant genetic engineering was considered as the most effective and economical means to bring up salt-tolerant plants and to exploit salt wasteland. The lack of efficient genes, salt-tolerant genes specially coming from halophytes has been the obstacle to limit its development. The main objectives of the dissertation were to utilize a cDNA library of halophyte Limonium sinense to exploit new efficient genes.High salt make the biomass of plants descended;Changeing the content and distributing of Na+ and K+ ;Some infiltrate adjust substances and MDA content changing.The nature of the damage that high salt concentrations inflict on plants is not entirely clear.An important cause of damage might be reactive oxygen species(ROS) generated by salt stress. To protect themselves against these toxic oxygen intermediates,plants employ defense systems that include the enzymes such as superoxide dismutases(SOD),catalases(CAT), peroxidase(POD),that catalyze the scavenging of ROS.1. The expression of k+-channel gene of Limonium sinenseThe seedlings of Limonium sinense were cultured in Hoagland with different salinities-0,100,200,300,400,500and 600 mmol.L- NaCl respectively. Northern analysis revealed the expression of k+-channel gene of Limonium sinense decrease then increase and then decrease with the increasing of NaCl concentration and it reaches the minimum under condition of 200 mmol.L- NaCl,The expression is maximum under condition of 300and 400 mmol.L- NaCl. 2.The expression of FLA gene of Limonium sinenseThe seedlings of Limonium sinense were cultured in Hoagland with 200 mmol.L- NaCl salinities. The times are 0,12,24,48,72,96 hours respectively.Northern analysis revealed the expression of FLA gene of Limonium sinense increase and then decrease with the increasing of times and it reaches the maximum under condition of 48 hours .3.The expression of NKCC gene of Limonium sinenseThe seedlings of Limonium sinense were cultured in Hoagland with 200 mmol.L- NaCl salinities. The times are 0,1,3,6,12,24 hours respectively.Northern analysis revealed the expression of FLA gene of Limonium sinense decrease with the increasing of times and it reaches the maximum under condition of 1 and 3 hours .4.Effects NaCl treatments on growth of Limonium sinenseThe growth of Limonium sinense decrease with the increasing of NaCl concentration and it reaches the maximum under condition of 100 mmol.L- NaCl,It is better than control and others are opposition.5.Effects NaCl treatments on Na+,K+,Cl- concentration of Limonium sinenseThe Na+ concentration of shoot and root of Limonium sinense increase with the increasing of NaCl concentration ; K+ concentration reaches the maximum under condition of 100 mmol.L- NaCl, It decrease with the changing of NaCl concentration ; Cl- concentration increase with the increasing of NaCl concentration.6.Effects NaCl treatments on MDA,SS,SOD,POD,CAT concentration of Limonium sinenseThe concentration of MDA is minimum under condition of 100 mmol.L- NaCl, It increase with the increasing of NaCl concentration and the concentration of SS opppsitionally; The concentration of SOD,CAT is maximum under condition of 100 mmol.L- NaCl, It decrease with the increasing of NaCl concentration;The activity of POD decrease with the increasing of NaCl concentration under condition of 100~500 mmol. L- but it increase distinctly under condition of 600 mmol.L- NaCl.
