By using of the technique of l)iffcrential Display RT-PCR (DDRT-PCR, the di lfercntiai expressIon pattern betwecn salt' treated plant and thc control were anaii.zecl. 330 differential displaycd cDNA fragments were isolated. Among them there were 60 fragments can he reampf i Vied by BCR. Half of these fragments had been used as probes on Northern hybridization to confirm the di fforcnces in l.he part. i ciii ar tnRNA I eve 1 s of salt-treated and untreate(l plants. ie results showed that the No. 136 cDNA fragment related transcript is highly induced when the plant exposures to high salt. This fragment was cloned and sequenced. The results of BLAST i.n GenBank hioinforinatic (ldtahases showed that the No. 136 fragment was similar to some genes for photosynthesis. We used the No. 136 fragment as probe to hybridize with more than 30, 000 phage clones of a uam[)da gtlO library constructed from poiy(A)~+RNA isolated from 48 hours NaCI (200mM) treated leaves from L. sinense. Several potential. phage clones were got and need further character i zat ion.
