To understand genetic condition of domestic Mongolian gerbil populations, the method of biochemical markers was established and the genetic structure and differentitation were studied in Mongolian gerbil populations, which are the biggest in our country. The study will provide theory foundation for breeding and preserving of gerbils, and technical means to gentic monitoring of closed gebil colonies; moreover, it has important significance in establishing genetic quality criterion of gerbil, and developing study and application of gebils.According to the biochemical markers method of rats and mice, 27 polymorphism-rich biochemical markers and 9 target tissues had been screened out, and best electrophoresis condition of gerbil had been optimized. The biochemical markers method had been elementarily established and applied to population genetic structure analysis. Analysing genetic diversity of four gerbil colonies by the method, the result showed that two gerbil colonies of Capital Universety of Medical Science had high level of genetic diversity and low degree of genetic similarity; the two colonies showed significantly difference in genetic adaptability, genetic diversity and genetic variation(P<0.05), compared with the original colony, the biological purifying colony had 7.14% gene lost, but the two colonies did not appear obvious genetic differentiation(FsT<0.05). The entire population had abundant genetic diversity in which the heterozygosity mean value was 0.1722, and the population deviated from Hardy-Weinberg equilibrium(P<0.01). Two gerbil colonies of Zhejiang Laboratory Animal Center also had high level of genetic diversity and low degree of genetic similarity; the two colonies showed significantly difference in genetic adaptability, genetic diversity and genetic variation(P<0.05), in contrast to the original colony, the biological purifying colony had 17.86% gene lost. The entire population had abundant genetic diversity in which the heterozygosity mean value was 0.1412, and the population deviated from Hardy-Weinberg equilibrium(P<0.05), but the population appeared obvious genetic differentiation (Fst<0.05).
