Research On The Regularity Of Clinical Medication For Precocious Puberty Based On Data Mining

Objective:By mining the data on the clinical use of traditional Chinese medicines for the treatment of precocious puberty basing on the theory of "correspondence between prescriptions and syndromes",this dissertation is to explore the etiology,pathogenesis,and the rules of syndromes for this disease.The data mining of Professor Ye Jin’s medical records of treating children’s precocious puberty is carried out to summarize her clinical experience and academic views for treating precocious puberty.The network pharmacology of the main prescription is analyzed to explore its mechanism and rationality.Methods:1.Collect 159 clinical research literatures on precocious puberty,and select 178 prescriptions to establish a database,and then use statistical methods including frequency,clustering,and association rule to analyze precocious puberty.2.According to 18 randomized controlled clinical research papers on precocious puberty were selected to conduct a Meta-analysis of the efficacy of traditional Chinese medicine in the treatment of precocious puberty.3.Collecting the medical precocious puberty records of Professor Ye Jin,using 245 cases of 745 medical records as the research object to establish a database,and then carry out data sorting and statistical analysis,use mining methods like frequency,clustering,and association rule mining to summarize the rule of medication for precocious puberty.4.Select Professor Ye Jin’s main prescription for the treatment of precocious puberty in children for network pharmacology research.Use TCMSP,Gene Card,OMIM,STRING database,Go and KEGG and R software.Result:1.Literature data mining researchDrug use:A total of 159 documents meeting the requirements were included,and 178 prescriptions were collected.There was a 142 kinds of Traditional Chinese Medicine(TCM)were involved,with a total frequency of 1664 times.According to the analysis of the efficacy of the herbs,the prescriptions for treating precocious puberty involved heat clearing herbs(37.19%),tonic herbs(17.915%),dampness-removing herbs(9.7%),qi-regulating herbs(7.2%),expectorant herbs(6.6%);Cold medicines accounted for 51.9%,cooling drugs accounted for 20.1%,bitter taste accounted for 37.66%,sweet taste accounted for 26.66%;the total proportion of drugs for eliminating pathogenic heat accounted for 63.795%,and the total proportion of drugs for nourishing yin accounted for 50.25%;The frequency was more than 35 times,and the cumulative drug frequency Fa>58.41%had a total of 14 drugs,namely Anemarrhena(65.17%),Cortex Phellodendri(62.29%),Cortex Moutan(55.05%),Rehmanniaglutinosa(47.19%),Poria(44.38%)),Prunella vulgaris(38.20%),Alisma(37.07%),Bupleurum(36.51%),Chinese yam(28.65%),Cornus(22.47%),Turtle(21.91%),Licorice(21.91%),Turmeric(21.34%)),tangerine peel(20.22%);Through cluster analysis and association rule mining,it was found that the common compatibility combinations of clinical medication for precocious puberty were Bupleurum,gentian,white peony,Prunella,oyster;turmeric,fritillary,gardenia,peony;Rehmanniaglutinosa;Tortoise shell,Rehmanniaglutinosa,Phellodendronrhizoma,Alisma;Poria,licorice,raw malt,Chinese yam;Pinellia;The meridian of the herbs showed that the kidney(20.48%),liver(19.72%),heart(12.68%),lung(12.61%),spleen(9.48%),stomach(12.57%).2.Literature Meta analysis results:Chinese medicine for precocious puberty girls has the effect of reducing the levels of FSH,LH and E2,inhibiting ovarian and uterine development,and delaying bone growth in girls with precocious puberty.3.Professor Ye Jin’s real-world therapeutic case analysis and study of precocious pubertyDrug use:714 prescriptions involved 89 kinds of traditional Chinese medicines,and the total frequency of use was 8864 times.From the view of the efficacy of the herbs,the prescriptions for the treatment of precocious puberty involved heat-removing herbs(43.44%),nourishing herbs(18.50%),blood-activating herbs(10.75%),expectorants herbs(7.97%),and dispelling herbs(5.76%);70.83%of the drugs are cold herbs,1.23%of cool herbs,40.26%of bitter and 23.65%of sweet;the frequency of occurrence is more than 100 times,and the cumulative drug frequency Fa>86.14%there are 16 drugs in total,respectively Anemarrhena(93.55%),Treats(91.73%),Scrophularia(86.97%),Turmeric(86.83%),Vinegar Tortoiseshell(85.15%),Fried White Peony(81.62%),Seaweed(79.69%),Prunella(79.41%),raw Rehmanniaglutinosa(75.21%),raw malt(64.42%),Cortex Moutan(57.56%),vinegar Bupleurum(56.30%),tangerine peel(43.97%),comfrey(42.15%),red peony(26.89%)Pinellia sinensis(15.40%);Through cluster analysis and association rule analysis,it was found that the common clinical compatibility combinations of YE Jin’s treatment of precocious puberty included comfrey,tangerine peel;Bupleurum,raw malt;Chuanxiong,fried Chunpi,French pinellia,red peony,Poria;Prunella vulgaris,turmeric,seaweed,Fried white peony root,Anemarrhena,salt Cork,Scrophularia,vinegar tortoise shell,peony bark;The meridian of the drug showed that the liver(24.56%),the kidney(20.13%),the lung(12.33%),the heart(11.94%),and the stomach(12.55%).4.Network Pharmacology Research and Molecular DockingAfter screening,89 active ingredients in the main prescriptions for precocious puberty could act on 91 targets of precocious puberty.The core targets involved ADRB2,PTGS2,etc.GO enrichment analysis yielded 125 entries,and KEGG enrichment analysis yielded 160 pathways.Conclusion:(1)The timely arrival of Tiangui is closely related to the stimulation,regulation and restriction of children’s physiological fire on reproductive activities.Various pathogenic factors lead to the excessive fire at the gate life,hyperactivity of kidney yang,full of essence and Qi in the kidney,resulting in the early arrival of Tiangui,which is the pathogenesis of the disease.The treatment of this disease is mainly based on kidney,liver,heart and spleen.(2)Traditional Chinese medicine is effective,safe and reliable in the treatment of precocious puberty in girls.(3)The data mining research results of Professor Ye Jin’s treatment of clinical medication further revealed that children with precocious puberty is associated with the new view of early arrival of Tiangui,due to insufficient endowment,juvenile Yin,vulnerable to external evil Yin injury,Yin deficiency life door fire prosperous,and excessive Wen Xi.Therapies for precocious precocity included clearing heat,nourishing Yin and strengthening water,cooling blood and nourishing blood,relieving depression,strengthening spleen and replenishing qi,and reducing phlegm and regulating kidney qi by clearing damp.(4)The regulation of precocious puberty by Ye Jin’s zhufang is regulated by multiple components,multiple targets and multiple pathways at the same time.Among them,the E2/ER estrogen signaling pathway,the EGFR/PI3K/Akt signaling pathway,β-End/μ/Gi/o signaling pathway inhibit the level of sex hormones;the βEnd/μ/Gì/o/GIRK signaling pathway,which regulates the β-endophenol peptide,inhibits the release of GnRH,and achieves the clinical efficacy of Tiangui slow to.
