Purpose: To analyze the traditional Chinese medicine formula and medication rule for obesity based on data mining and to explore the potential mechanism of action of high-frequency drugs for the treatment of obesity by network pharmacology method.Data and methods: Chinese National Knowledge Infrastructure(CNKI),China Biology Medicine disc(CBMdisc),VIP and Wangfang Data were used as the data sources.Frequency analysis,association rules analysis,cluster analysis and principal component analysis(PCA)were carried out on these data by using Excel 2016 and statistical software R3.5.2,and the compounds contained in high-frequency drugs were searched by using BATMAN-TCM system to obtain the targets corresponding to these compounds.These targets were mapped to obesity-associated targets searched in Genecard database and intersection elements were obtained for network topology analysis by building a PPI network based on STRING 11.0 to screen out the core targets of high-frequency drugs for the treatment of obesity.KEGG pathway was analyzed through DAVID 6.8 Database to obtain the important pathways of high-frequency drugs for obesity.Results:1.A total of 143 literatures meeting the exclusion criteria were selected,and traditional Chinese medicine formulas and 172 drugs were included.2.The statistical results of the frequency of traditional Chinese medicine show that there are21 kinds of drugs with a frequency of drug use ?10%,among which the top five are Poria(56.75%),Alisma(50%),Atractylodes(43.93%),hawthorn(39.86%),Pinellia(27.03%).3.The results of the association rules show that the drug association rules are sorted by 6drug combinations in order of support,confidence,and promotion,and the drug pairs ranked from highest to lowest are Atractylodes and Poria,Banxia and Poria,Chenpi and Poria,Atractylodes and Poria,Alisma,Atractylodes and Poria,Hawthorn,Atractylodes and Poria.The drug pairs sorted from high to low are Licorice,Pinellia and Poria,Pinellia,Atractylodes and Poria,Pinellia,Citrus and Poria,Pinellia,Alisma and Poria,Atractylodes,Atractylodesand Poria,Chenpi Atractylodes macrocephala and Poria.The medicine pairs sorted from high to low are orange peel,Poria and Pinellia,Alisma and Dan participate in hawthorn,Dan participates in Hawthorn,Poria,Alisma and Dan participate in Atractylodes,Licorice,Pinellia and Poria,Pinellia,Atractylodes macrocephala and Poria.4.Cluster analysis results show that there are two clustered drug groups: the first group contains drugs: Alisma,Poria,Atractylodes,hawthorn,lotus leaf,Salvia,Pinellia,orange peel,Atractylodes,Magnolia,Coix Kernel,astragalus,licorice.The representative group of the second group of drugs mainly includes: rhubarb,citrus aurantium,Bupleurum,white peony root,ginger,jujube,scutellaria baicalensis,Coptis chinensis.5.The principal component analysis results show that the principal components of the first four dimensions are presented.Among them,the medicines in dimension 1 mainly include:Poria,Atractylodes,Rhubarb,Atractylodes,Pinellia,Alisma,etc.,and the medicines in dimension 2 include: Hawthorn,cassia,lotus leaf,Alisma,salvia,etc.,the main medicines included in dimension 3 are: Pinellia,orange peel,astragalus,hawthorn,citrus aurantium,Fang Ji,Magnolia,etc.,the main medicines included in dimension 4 are: Atractylodes macrocephala Diarrhea,licorice,herb,hawthorn,salvia,cassia and so on.6.The results of high-frequency drugs Poria and obesity-related targets showed that the core targets of Poria in the treatment of obesity were found through network pharmacology methods ANXA1,HTR1 A,CXCR4,ADRA2 B,CNR1,ADRA2 A,HTR1B,PTGER3,DRD2,etc.There are 20 important signal pathways such as serotoninergic synapse,TRP channel inflammatory mediators,adipokine signaling pathway,insulin resistance,insulin signaling pathway and so on.Conclusion:1.In traditional Chinese medicine,the treatment of obesity focuses on the regulation of “qi deficiency”,“phlegm-dampness” and “blood stasis”.2.Chinese medicine treatment of obesity is based on strengthening the spleen and dampening the phlegm,so the medication is mostly Poria,Atractylodes,Citrus aurantium,Atractylodes,Astragalus,Coix,Pinellia,Thick Park and others are mainly supplemented by promoting blood circulation,removing blood stasis,diarrhea,hawthorn,salvia,lotus leaf,etc.3.3.ANXA1,HTR1 A,CXCR4,ADRA2 B,CNR1,ADRA2 A,HTR1B,PTGER3,DRD2,DRD3 and other important pathways such as serotoninergic synapse,TRP channel inflammation mediator regulation,fat cells Factor signaling pathway,insulin resistance,insulin signaling pathway,etc.can provide a reference for further research on the mechanism of Poria's treatment of obesity.
