Research purpose: Based on clinical case data to comprehensively explore Song Ligong’s clinical treatment of insomnia ideas,medication rules and explore the core prescription,and apply network pharmacology to explore the mechanism of high-frequency syndrome type core prescription in the treatment of insomnia,so as to provide objective reference for the treatment of insomnia with traditional Chinese medicine,so as to better guide the clinical.Research methods:(1)Based on the electronic medical record data of all patients received by Mr.Song Ligong from January to December 2020 as the database,according to the inclusion and exclusion criteria,the corresponding medical records diagnosed as insomnia by TCM were selected,and the drugs and symptoms of the medical records were standardized,and entered into the TCM inheritance assistance platform V2.5.The auxiliary platform of traditional Chinese medicine inheritance is used to carry out statistical analysis on the general situation of patients,drug use frequency,sex and taste,meridian tropism,and drug use frequency of main syndrome types,cluster analysis and prescription rule analysis on the included prescriptions,and entropy clustering of core drug combinations and new prescriptions,so as to explore the medication rule of Song Ligong’s treatment of insomnia.(2)TCMSP,Pubchem and other databases are used to collect the effective active ingredients of the core prescription of the high-frequency syndrome type(liver-qi stagnation and fire-transforming syndrome)and predict its target,and the Genecards database is used to search with "insomnia" to collect the insomnia related targets,and the obtained disease target and the core prescription target are mapped through the Venny 2.1.0 platform to obtain the intersection target,and the "drug-effective componenttarget" network is further constructed using Cytascape 3.8.2.The obtained intersection targets are screened by constructing protein interaction(PPI)network through STRING platform and Cytascape 3.8.2.Through the analysis of KEGG pathway and gene ontology(GO)enrichment through Metascape database,explore the potential biological process and signal pathway of core prescription for insomnia treatment.Research results:(1)Data mining results: Of the 285 cases of insomnia,154 were female insomnia patients,accounting for 64.56%;101 male patients with insomnia,accounting for 35.44%,male: female=1:1.82;There are 46 kinds of symptoms involved in the included patients,and the highest frequency is respectively upset,fatigue,difficulty in falling asleep,dizziness,dreaminess,early awakening,irritability,anxiety,easy awakening,etc;A total of 386 prescriptions were collected and 165 kinds of traditional Chinese medicine were used,with a total frequency of 4569 times.The first60 drugs with the highest frequency were used 4086 times,accounting for89.42% of the total frequency.There were eight traditional Chinese medicine with a frequency of more than 50%,including Coptis chinensis(63.64%),Poria cocos(62.94%),Bupleurum chinense(61.54%),Polygonum multiflorum(58.74%),Angelica sinensis(55.59%),Semen ziziphi spinosae(54.20%),Radix paeoniae alba(53.50%)and Polygala(51.40%);The four qi of traditional Chinese medicine are mainly cold,warm and flat,with the frequency of 35.98%,32.03% and 26.76% respectively,and the use of cold and hot drugs is less;Among the five flavors of traditional Chinese medicine,the most are sweet,bitter and bitter,accounting for 33.97%,29.86% and 23.77% respectively.The use of sour,astringent and salty drugs is less;Among all the prescription drugs,the drugs that belong to the heart channel have the highest frequency,2255 times,accounting for 17.57%,followed by the liver channel,2050 times,accounting for 15.98%;Commonly used drug compatibility: Bupleurum chinense-Poria cocos,Rhizoma coptidis-Bupleurum chinense,Rhizoma coptidis-Cauliflower,Radix scutellariae-Bupleuri,Rhizoma coptidis-Poria cocos.The new formula of clustering is: 1.Muxiang-longan meat-atractylodes macrocephala-astragalus-keel;2.Orange peel-Angelica sinensis-Magnolia officinalis-Zhuru-Pinellia ternate-Fructus aurantii;3.Forsythia suspensa-honeysuckle-black ginseng-light soybeans-lotus seed heart-mother of pearl-light bamboo leaves;4.bitter almond-wormwood-cardamom-gentian;5.Fushen-Polygala tenuifolia-Acorus calamus-Lily-Cypress seed;6.Gardenia jasminoides-Chuanxiong-Xiangfu-Shenqu-Cangshu;7.Rehmannia glutinosa-Chinese yam-cornus officinalis-alisma orientalis-peony skin-scutellaria baicalensis-bupleurum chinense-oyster-keel-cinnamon twig.(2)Network pharmacological analysis results: 93 compounds and 273 action targets were obtained after screening.After constructing the "drug effective compound disease target" interaction network,quercetin β-The PPI interaction network analysis of important active ingredients such as sitosterol,kaempferol,stigmasterol,baicalein,and other key targets such as VEGFA,IL6,TNF,AKT1,CASP3.Using Metascape database to conduct GO analysis and KEGG analysis on 126 targets,4774 BP entries,427 CC entries and 681 MF entries were obtained.A total of 252 signal pathways were obtained by KEGG enrichment analysis.The main pathways that play a role may be calcium signal pathway,5-hydroxytryptaminergic synapse,dopaminergic synapse and other signal pathways.Research conclusion:1.Use the method of data mining to explore the medication rule of Mr.Song Ligong’s treatment of insomnia.The core drugs are Coptis chinensis,Poria cocos,Bupleurum chinense,Polygonum multiflorum,Angelica sinensis,Semen ziziphi spinosae,Radix paeoniae alba,Radix Polygalae,etc.2.The core prescription may pass quercetin β-The important compounds such as sitosterol,kaempferol,stigmasterol,baicalin act on the core targets such as VEGFA,IL6,TNF,AKT1,CASP3,to regulate the signal pathways such as calcium signaling pathway,5-hydroxytryptaminergic synapse,dopaminergic synapse,and play its therapeutic role.
