Professor Qi Yuanfu’s Clinical Experience In The Treatment Of Lung Cancer And The Experimental Study On Regulating Main Targets Of PI3K/AKT Pathway By Common Heat Clearing And Detoxifying Drugs

Objective: Through data mining technology,this article studied Professor Qi Yuanfu’s prescription drugs from multiple levels and angles,and summarized relevant medication rules and clinical experience in the treatment of lung cancer.Common heat-clearing and detoxicating drugs used by Professor Qi Yuanfu were extracted for in vitro antitumor experimental research.Methods: The first part was the study of medical records.By collecting the prescriptions of Professor Qi Yuanfu’s of lung cancer,we analyzed the pathological types,symptoms,tongue and pulse,pathogenesis and drug taste of patients,cluster and relevancy rule analysis,to sum up Professor Qi Yuanfu’s prescription rules and to talk over Professor Qi Yuanfu’s clinical experience.In the second part,the drug containing serum of Professor Qi Yuanfu’s commonly used antipyretic and detoxifying drugs was prepared,and the cells were cultured routinely.CCK-8 was used to detect the changes of multiplication potentiality of BEAS-2B,A549 and H1299,so as to clarify the toxic effects of targeted drugs on different cells,and it is convenient for showing the toxicity of the object drugs on distinct cells.The function of object drugs on the migration and invasion of A549 and H1299 cells and the adjusting of PI3K/AKT pathway correlative target spots were found out by scratch test,Transwell invasion experiment,PCR and WB.Results:(1)In the medical record research,there were 610 patients who were included.The main pathological types were lung adenocarcinoma(48.85%)and lung squamous cell carcinoma(32.95%).The most common symptoms of lung cancer were cough,followed by fatigue,expectoration,poor sleep,shortness of breath,weight loss,loose stools,etc.The tongue is light red,dark purple,thin and yellow fur are the most common,and the frequency of fine veins and stringed veins is the highest.The drug taste is mainly mild,and the meridians of lung,spleen,stomach and liver are the most.The commonly used antipyretic and detoxifying drugs are flea rest,Scutellaria baicalensis and Hedyotis diffusa,snakeberry and Baiying,which can help healthy qi,such as Ligustrum lucidum and dodder,Astragalus membranaceus and Pseudostellaria Pseudostellariae,Atractylodes macrocephala and Fructus aurantii,resolving phlegm,relieving cough and relieving asthma,such as Platycodon grandiflorum and almond,white front and Peucedanum,aster and winter flower,Chenpi and Pinellia ternata.(2)In the experimental part,Professor Qi Yuanfu’s commonly used antipyretic and detoxifying drugs(flea rest,Hedyotis diffusa,Scutellaria baicalensis,snakeberry and Baiying)serum has no obvious toxic effect in normal lung epithelial cells BEAS-2B,and has a concentration dependent obvious toxic effect in NSCLC(A549,H1299)cell lines,which can inhibit the proliferation,migration and invasion of two kinds of lung adenocarcinoma cells,Combined with cisplatin has synergistic effect.it can lower the gene and protein expression levels of PI3 K and AKT and adjust upward the expression level of PTEN protein,which may restrain the development of neoplasm.Conclusion:(1)Based on the statistics of medication law,it is found that interpretation of the cause,onset and process of lung cancer patients is the combination of phlegm,heat and toxin and the deficiency of Qi and Yin.The disease is located in the lung and involves lung,stomach,liver,kidney.(2)Professor Qi Yuanfu’s treatment principle for lung cancer is to support the right and dispel the evil,attack and supplement at the same time.On the basis of paying attention to both qi and Yin,he advocates that clearing away the heat and resolving phlegm should be used to dispel the evil.In the prescription,there are more drugs for dispelling evils than for strengthening health.Generally speaking,"Dispelling evils is greater than strengthening health".At the same time,it is concluded that the basic core formula of heat clearing and detoxifying drugs is composed of five drugs,namely: flea rest,Hedyotis diffusa,Scutellaria baicalensis,snakeberry and Baiying.It supports healthy qi,such as Ligustrum lucidum and dodder,Astragalus membranaceus and Radix Pseudostellariae,Atractylodes Macrocephalae and Fructus aurantii,resolving phlegm,relieving cough and relieving asthma,such as Platycodon grandiflorum and almond,Baiqian and Qianhu,aster and winter flower,tangerine peel and Pinellia ternata.Qingre Jiedu therapy has a good inhibitory effect on lung cancer,which provides more choices for clinical treatment of lung cancer.(3)Taking Professor Qi Yuanfu’s basic core prescription of heat clearing and detoxifying drugs commonly used in the treatment of lung cancer as the starting point,In vitro experiment have illustrated that the commonly used Qingrejiedu drugs can inhibit the proliferation,migration and invasion of A549 and H1299 cells,and its mechanism may be relevant to the main target of regulating PI3K/AKT pathway.
