Purpose:To observe Weitong Xiaopi prescription the change of behavior,gastric residual rate,propelling rate of small intestine,ghrelin related receptors in gastric antrum tissue and hypothalamus in Functional dyspepsia rats.To explore the possible mechanism of Weitong Xiaopi prescription on the FD rats from brain-gut axis and brain-gut peptide,providing a new theoretical basis for traditional Chinese medicine to treatment functional dyspepsia,and provide a theoretical and experimental basis for the development of new drugs.Material and method:48 healthy SD SPF rats were randomly divided into 4 groups:the blank control group,the model control group,the Weitong Xiaopi prescription group and the Mosapride group,12 rats in each group.Beside the blank control group,the other groups were used Guo clip tail stimulation combined with Zhang irregular feeding method 21 days to create rat model.After the FD rats model were established successful,to observe the general condition and behavior.During the period of treating,the blank control group and model group were given intragastric administration of normal saline.The Weitong Xiaopi prescription contains: Dangshen 10 g,Caihu 12 g,Baishao 20 g,Yanhusuo 20 g,Baizhu 20 g,Sharen 15 g,Zhishi 12 g,Caomaiya 20 g,Zhigancao 6g,Jiaoshanzha 10 g,Jineijin 10 g.Mosapride citrate tablets were used as western medicine control.The Weitong Xiaopi prescription group and the Mosapride group were given granule of Weitong Xiaopi prescription and Mosapride tablets diluted solution according to the body surface area conversion method between human and rat,twice a day,continuous administration 21 days.21 days later,observe the general condition score.The rats were fasted but drinking freely 24 hours,all groups were fed with nutritive semi-solid paste mixed with ink.The rats of all groups were subjected to the abdominal anesthesia in turn and were killed.Gastric residual rate,propelling rate of small intestine were observed.5-HT and BDNF in gastric antrum tissue and hypothalamus were measured by IHC.The gene and protein expression of BDNF,5-HT3aR,a-CGRP,GPR39 in gastric antrum tissue and hypothalamus were measured by RT-PCR and WB.Results:1.The general condition of ratsAfter FD model was established,the blank control group rats have shiny coat,good mental state,good activity,normal dieting and normal defecation,higher bodyweight,muscular build,normal excretion,and no death.The model group rats were no shiny coat,poor mental state and activity,decrease diet,slow weight gain or loss.After 3 weeks treatment,the general condition and behaviorof model group were bad,and the Weitong Xiaopi prescription group and the Mosapride group rats were better,but have no significant difference.8 rats were died during the experiment.2.Gastrointestinal motility change of rats in all groupsCompare with the blank control group,the propelling rate of small intestine of the model group was decreased(P<0.05).Compare with the model group,the propelling rate of small intestine of the Weitong Xiaopi prescription group and the Mosapride group were increased(P<0.05),and the Weitong Xiaopi prescription group was better than the Mosapride group(P<0.05).Compare with the blank control group,the gastric residual rate of the model group was increased(P<0.05).Compare with the model group,the gastric residual rate of the Weitong Xiaopi prescription group and the Mosapride group were decreased(P<0.05),and the Weitong Xiaopi prescription group was better than the Mosapride group(P<0.05).3.The expression of 5-HT and BDNF in the gastric antrum tissue and hypothalamus Compare with the blank control group,the expression of 5-HT and BDNF in the gastric antrum tissue and hypothalamus of the model group was decreased by immunohistochemistry(P<0.05).Compare with the model group,the expression of 5-HT and BDNF in the gastric antrum tissue and hypothalamus of the Weitong Xiaopi prescription group and the Mosapride group were increased(P<0.05),and the Weitong Xiaopi prescription group was better than the Mosapride group(P>0.05).4.The expression of BDNF,5-HT3aR,a-CGRP,GPR39 mRNA in the gastric antrum tissue Compare with the blank control group,the expression of BDNF,5-HT3aR,a-CGRP mRNA in the gastric antrum tissue of the model group was decreased(P<0.05).Compare with the model group,the expression of BDNF,5-HT3aR,a-CGRP mRNA in the gastric antrum tissue of the Weitong Xiaopi prescription group and the Mosapride group were increased(P<0.05),and the Weitong Xiaopi prescription group was better than the Mosapride group(P<0.05).Compare with the blank control group,the expression of GPR39 mRNA in the gastric antrum tissue of the model group was increased(P<0.01).Compare with the model group,the expression of GPR39 mRNA in the gastric antrum tissue of the Weitong Xiaopi prescription group and the Mosapride group were decreased(P<0.05),and the Weitong Xiaopi prescription group was better than the Mosapride group(P<0.05).5.The expression of BDNF,5-HT3aR,a-CGRP,GPR39 mRNA in the hypothalamus Compare with the blank control group,the expression of BDNF,a-CGRP,5-HT3aR mRNA in the hypothalamus of the model group was decreased(P<0.05).Compare with the model group,the expression of BDNF,5-HT3aR,a-CGRP mRNA in the hypothalamus of the Weitong Xiaopi prescription group and the Mosapride group were increased(P<0.05),and the Weitong Xiaopi prescription group was better than the Mosapride group(P<0.05).Compare with the blank control group,the expression of GPR39 mRNA in the hypothalamus of the model group was increased(P<0.01).Compare with the model group,the expression of GPR39 mRNA in the hypothalamus of the Weitong Xiaopi prescription group and the Mosapride group were decreased(P<0.05),and the Weitong Xiaopi prescription group was better than the Mosapride group(P>0.05).6.The expression of BDNF,5-HT3aR,a-CGRP,GPR39 protein in the gastric antrum tissue Compare with the blank control group,the expression of BDNF,a-CGRP protein in the gastric antrum tissue of the model group was decreased(P<0.05),the expression of 5-HT3aR protein in the gastric antrum tissue of the model group was decreased(P<0.01).Compare with the model group,the expression of BDNF,5-HT3aR,a-CGRP protein in the gastric antrum tissue of the Weitong Xiaopi prescription group and the Mosapride group were increased(P<0.05),and the Weitong Xiaopi prescription group was better than the Mosapride group(P<0.05).Compare with the blank control group,the expression of GPR39 protein in the gastric antrum tissue of the model group was increased(P<0.01).Compare with the model group,the expression of GPR39 propteom in the gastric antrum tissue of the Weitong Xiaopi prescription group and the Mosapride group were decreased(P<0.05),and the Weitong Xiaopi prescription group was better than the Mosapride group(P<0.05).7.The expression of BDNF,5-HT3aR,a-CGRP,GPR39 protein in the hypothalamus Compare with the blank control group,the expression of BDNF,5-HT3aR protein in the hypothalamus of the model group was decreased(P<0.05),the expression of a-CGRP protein in the hypothalamus of the model group was decreased(P<0.01).Compare with the model group,the expression of BDNF,5-HT3aR,a-CGRP protein in the hypothalamus of the Weitong Xiaopi prescription group and the Mosapride group were increased(P<0.05),and the Weitong Xiaopi prescription group was better than the Mosapride group(P<0.05).Compare with the blank control group,the expression of GPR39 protein in the hypothalamus of the model group was increased(P<0.01).Compare with the model group,the expression of GPR39 protein in the hypothalamus of the Weitong Xiaopi prescription group and the Mosapride group were decreased(P<0.05),and the Weitong Xiaopi prescription group was better than the Mosapride group(P>0.05).Conclusion:1.Guo clip tail stimulation combined with Zhang irregular feeding method can replicate FD rats model.2.Weitong Xiaopi prescription can improve the color of FD rats,improve activities,increase food intake,increase body weight,and relieve irritability and irritability behavioral symptoms,thus proving that Weitong Xiaopi Recipe has a therapeutic effect on FD.3.Weitong Xiaopi prescription reduce the gastric residual rate of FD rats,increase the intestinal propulsion rate,and improve gastrointestinal motility is one of the mechanisms of Weitong Xiaopi prescription in the treatment of FD.4.The mechanism of Weitong Xiaopi prescription in the treatment of FD is to increase the expression of 5-HT and BDNF and treat FD from the aspect of mental and psychological factors,which is one of its mechanisms.5.There are BDNF,5-HT3aR,a-CGRP,GRP39 mRNA brain-gut interaction positive feedback mechanism in the treatment of FD with Weitong Xiaopi prescription.6.Weitong Xiaopi prescription can increase the expression of 5-HT,BDNF,?-CGRP,and reduce the expression of GPR39 by BGA to treat FD.
