Objective:From the point of view of synaptic plasticity,this paper discusses the molecular mechanism of Wuzang Wenyang Huayu Decoction to protect and repair the learning and memory ability of VD rats,and explains Professor Tang Nong's theory of Yang deficiency and phlegm stasis pathogenesis of vascular dementia and its clinical significance.Method:In this paper,theoretical and experimental research methods are used.1.Theoretical research:by combing the general situation of modern medical research and traditional Chinese medicine research of VD,reviewing Professor Tang Nong's school inheritance and academic views,this paper explains Professor Tang Nong's understanding of the etiology and pathogenesis of vascular dementia and its guiding significance for treatment.2.Experimental research:The VD model of SD rats with chronic cerebral ischemiawas made by ligation of bilateral common carotid arteries.The Morris water maze score of one week after operation was used as the criteria for the success of the model.Taking sham operation as negative control and nimodipine as positive control,Wuzang Wenyang Huayu decoction was given for 4 weeks.Observe the effect of Wuzang Wenyang Huayu Decoction The effects of learning and memory ability,navigation and space exploration on VD rats were analyzed,and the abilities of learning and memory and space recognition were analyzed.The expression levels of synaptic plasticity related presynaptic membrane synapsin and growth promoting protein GAP-43 in hippocampus were detected by immunohistochemistry and Western blotting,and the synaptic plasticity related by Wuzang Wenyang Huayu decoction was detected by ELISA The changes of VEGF,BDNF and NGF in serum were studied to explore the protective mechanism of Wuzang Wenyang Huayu Decoction on synaptic plasticity in VD rats.Result:VD is based on the deficiency of Yuanyang,the stagnation of brain and collaterals by phlegm and blood stasis,and the deficiency and the excess.The treatment should be based on Warming Yang,removing blood stasis and eliminating phlegm.The mortality of vascular dementia model can be reduced by permanent ligation of bilateral common carotid artery.Through Morris water maze test,the model made by permanent ligation of bilateral common carotid artery can appear learning and memory disorders.One month after the operation,the model group was more timid and frightened,more curled up,less eyeball,lighter weight,less independent activity,soft or shapeless stool,low glossiness of hair,light color of claw nail,low compatibility with stomach perfusion,and easily irritated.Wuzang Wenyang Huayu Decoction group had a general mental state after operation,which was significantly improved after drug treatment,with rapid growth and development,more independent activities,stool formation,glossy hair,normal color of claw nail,and gastric perfusion.In Morris water maze navigation experiment,Wuzang Wenyang Huayu Decoction group can shorten the escape latency and the total distance to find the platform,reduce the time to cross the third quadrant for the first time,and increase the time to stay in the target quadrant in the space exploration experiment.This shows that Wuzang Wenyang Huayu Decoction can improve the long-term learning and memory ability of VD rats.In immunohistochemistry and Western blot experiments,Wuzang Wenyang Huayu Decoction group can increase the expression of Syn and GAP-43 protein in CA1 area of hippocampus,suggesting that Wuzang Wenyang Huayu Decoction can improve the synaptic plasticity of neurons by up regulating the expression of Syn and GAP-43 protein.The results of ELISA showed that Wuzang Wenyang Huayu Decoction group could improve the serum VEGF level of VD rats,suggesting that Wuzang Wenyang Huayu Decoction group could improve the synaptic microenvironment of neurons.Wuzang Wenyang Huayu Decoction group can improve the level of BDNF and NGF in serum of VD model rats,which shows that Wuzang Wenyang Huayu Decoction group can improve the condition of neuron synapse damage under the condition of chronic cerebral ischemia and hypoxia after operation.Wuzang Wenyang Huayu Decoction can improve the learning and memory behavior and synaptic plasticity pathology of hippocampus in VD rats.Syn,GAP-43,VEGF,BDNF and NGF are significantly higher than those before treatment.Conclusion:(1)The pathogenesis of vascular dementia is based on the deficiency of Yang Qi and the loss of Shenji.It takes the deficiency of Yang and the stagnation of brain collaterals by blood stasis and phlegm as the standard.Warming yang and activating blood circulation and resolving phlegm is one of the basic treatment methods for vascular dementia.The five zang organs Warming Yang and Removing Stasis Decoction warming yang and transforming Qi has a good effect on the treatment of vascular dementia.(2)Wuzang Wenyang Huayu decoction has the effect of improving the learning and memory ability of vascular dementia rats,highlighting the relationship between Yang Qi and spirit,which is consistent with the theory of "essence is nourishing spirit".(3)Wuzang Wenyang Huayu Decoction can improve the learning and memory ability of vascular dementia rats,which may be related to the regulation of synaptic plasticity of hippocampal neurons in vascular dementia rats,specifically in improving the expression of syn and GAP-43 in presynaptic membrane protein,as well as up regulating the levels of VEGF,BDNF and NGF in serum molecules related to synaptic plasticity.
