Objective:To explore the effects of Chaoyi Bawujunzi decoction on learning and memory ability and the morphology of hippocampal CA1 region.Method:In this study,50 male SD rats were randomly divided into sham operation group and operation group,and the operation group adopted the modified 2-vascular occlusion method(2-VO)makes dementia rat model,the sham operation group only separates without ligation.The successful rats were then randomly divided into five groups:the sham operation group,the model group,the nicergoline group,the high-dose group of the Chaoyi Bawujunzi decoction,and the low-dose group of the Chaoyi Bawujunzi decoction.On the next day after the operation,stomach was began to be filled,the sham operation group and the model group were given distilled water 10ml/kg/d,nicergoline group was given the nicergoline of 0.5 ml/kg/d,high-dose group was given the Chaoyi Bawujunzi decoction of 18 g/kg/d,low-dose group to the doctor eight things gentleman soup 4.5 g/kg/d,continuous lavage of the stomach was carried out for 28 d.Morris water labyrinth experiment was carried out 5 days before the end of gastric lavage to observe the changes of behavioral ability before and after treatment in VD rats.After death,he staining was used to observe the structural morphology of neurons in hippocampus CA1 region,and the changes of SOD,MDA and GSH-PX levels in serum of VD model rats were observed.Result:1.The experimental study of morris water labyrinth showed that compared with the model group,the incubation period of rats in the Chaoyi Bawujunzi decoction(high and low dose group)was significantly shortened,and the number of crossing platforms increased significantly.2.From he staining,it can be seen:in the sham-operated group,the morphology of hippocampal CAI region was not abnormal,the arrangement was orderly and the boundary was clear,and in the model group,the cell arrangement in the hippocampal CAl region was disorder,and the cell morphology variation and uneven staining were observed.In nicerjiaolin group,high dose group of Chaoyi Bawujunzii decoction and low dose group of Chaoyi Bawujunzi decoction,the shape of oval cells was increased and the arrangement was more regular than that of model group.3.Serum SOD,MDA,GSHH-PX:compared with the sham operation group,the content of SOD and GSH-PX in the serum of the model group was significantly reduced,the content of MDA was significantly increased(P<0.001).Compared with the model group,the levels of SOD,GSH-PX in serum of high dose group and low dose group of Chaoyi Bawujunzi decoction were higher and the content of MDA was lower than that of model group.Conclusion:1.The Chaoyi Bawujunzi decoction can significantly improve the learning and memory ability and space exploration ability of VD model rats.2.Chaoyi Bawujunzi decoction has a protective effect on the cell damage in the hippocampus CA1 region of VD model rats.3.Chaoyi Bawujunzi decoction can regulate VD model rat serum SOD,GSH-PX activity,reduce MDA content.
