Primary oral mucosal melanoma(OMM),a set of exceedingly rare neoplasm,is the most dismal and aggressive malignant tumor in the oral and maxillofacial regions. As a biologically distinct subtype of melanoma,OMM differed from cutaneous melanomas in histology and that the prognosis of OMM was poorer than cutaneous melanomas. However, due to their rarity,OMM remains poorly understood, calling for further investigation of its prognostic parameters and therapeutic effect of targeted therapy. The aims of the present study were to explore the useful predictive prognostic factor of OMM and provide a basis for the targeted therapy of metastatic OMM.In the present study,(1)we find out the status of cervical lymph node and the Ki67 expression were independent prognostic factor for OMM without distant metastases.Ki67 expression could act as an useful pathological predictor of survival of OMM patients without distant metastases;(2)For distant metastatic OMM,the status of primary lesion, positive cervical lymph node and c-KIT mutations were significant prognostic factors;(3)we found that Imatinib may benefit metastatic OMM patients with c-KIT mutations;(4)forthermore,our results indicated that the activity of m TOR was associated with Imatinib-resisdence in distant metastatic OMM.In conclusion, this research undertooka study on the predictive prognostic factor of OMM at different stage. The results build a foundation forprognosis system, provide the theory base for the use of targeted therapy and preliminary explore the protential candidate target of Imatinibresisdence.
