| Objective:Longjiang Han’s of gynaecologybased on the treatment of endometriosis(EMs)of academic thoughts and clinical experience,medication rule and in the cutting,in the observation of traditional Chinese medicine compound different analgesic soup of ectopic endometrium tissue pathology change of EMs rats and the effect of exploration of estradiol(E2),estrogen receptor(ER),aromatase P450 and nuclear transcription factors involved in regulating NF-κB and AP-1 EMs occurrence,development process,to clarify the traditional Chinese medicine ni yi zhi tong decoctions inhibition mechanism of ectopic endometrial hyperplasia,for traditional Chinese medicine compound with in ni yi zhi tong decoctions for new drugs for the treatment of EMs.Methods:1.By collect longjiang Han’s gynecological clinical basis and published papers and works for many years,to analyze asaps treatment of department of gynaecology EMs academic thought,dialectical thinking and drug laws,and in the cutting,in order to make clear of traditional ni yi zhi tong decoctions treatment effectiveness of the EMs.2.Autologous grafting EMs rats model,the selection of 57 only SD rats for autologous transplantation,and in a second caesarean section 4 weeks after surgery,observation of ectopic endometrial lesions form change,HE staining observation of ectopic endometrium tissue pathology change,evaluation of building was successful.Will building successful within the EMs only 40 rats were randomly divided into different analgesic decoction high dose group,the different analgesic soup low dose group,danazol positive drug group and model control group,each group of 10,only setting up control eight only.Groups of rats were given corresponding drugs and saline lavage,4 weeks after treatment to death and take the ectopic endometrium tissue and serum.3.By caesarean section and HE staining to observe each group rats of ectopic endometrium tissue pathological morphological changes after the treatment;Using enzyme-linked immunoassay(ELISA)to detect each group rats serum E2,and after the treatment of ectopic endometrium tissue of slurry in the ER and the change of aromatase P450,in order to make clear compound in different analgesic effect on the stability of the EMs disease soup,further by rt-pcr detection of c-Jun,c-Fox gene transcription,immune histochemical method to detect the c-Jun,c-Fox,the nf-kappaBp50,nf-kappaBp65 expression and protein imprinting method to detect the c-Jun,c-Fox and nf-kappaBp50,the expression of the nf-kappaBp65,explore compound within different analgesic mechanism of soup on the treatment of diseases in EMs.Results:1.Longjiang Han’s think endometriosis of department of gynaecology belong to Chinese medicine dysmenorrhea,remove lump,infertility,menstruation to be not moved,and other disease of department of gynaecology of integrated performance.Han’s the main use of traditional Chinese medicine theory,combined with clinical experience and modern testing method,the "disease",starting from the overall to local,the liver qi and remove stasis analgesia,founded "NI YI ZHI TONG DECOCTIONS",at the same time according to different individual dialectical,providing necessary combination of traditional Chinese medicine enema therapy,internal and external governance,curative effect.2.Autologous grafting EMs rats model building success rate is above 78%.Visible to the naked eye in the rat model of success the abdominal wall has a transparent vesicle formation,visible on the surface of fresh blood vessels,and violet bag piece,see local lesion and pelvic adhesion.HE dyed microscopically,normal endometrial stromal cells arranged closely,glandular structure clear,epithelial integrity.After building the ectopic endometrium of rats can be found that the number of mesenchymal cells increased,compact arrangement disordered,uneven distribution,deformation of glands atrophy,arrange irregular nucleus,ectopic endometrium tissue to form.3.Treatment of each group rats was observed 4 weeks after caesarean section,the treatment group organization cyst volume of endometriosis lesions than before treatment significantly narrowed,model group before and after treatment of endometriosis lesions organization cyst volume has no obvious change.HE staining pathology observation results show that compared with control decoctions of normal endometrium,model decoctions,uneven distribution of ectopic endometrial stromal cells and glands shrinking deformation,the nucleus arranged neatly;Traditional ni yi zhi tong decoctions in high and low dose decoctions and western medicine decoctions of ectopic endometrium and the control decoctions of normal endometrial pathological morphology is similar.4.Enzyme-linked immunoassay,according to the results of the rats after treatment,drug treatment group compared with model group,E2,ER and aromatase P450 content decreased obviously.Compared with the traditional Chinese medicine low dose group,high dose group of EMs model rats serum E2 and ectopic tissues ER and aromatase P450 content decreased significantly,the difference was statistically significant(P<0.05).5.PCR results showed that different analgesic decoctions after the intervention of traditional Chinese medicine compound,EMs model rats decoctions c-Jun,c-Fox gene transcription level significantly decreased(P<0.01);By using immunohistochemistry and Western blot method to detect the related proteins,the results show that traditional Chinese medicine compound within different analgesic decoctions can significantly reduce the EMs model of ectopic endometrium tissue of rats in c-Jun,c-Fox and nf-kappaBp50 and nf-kappaBp65 protein expression,and Western blot results show that the effect is more apparent,high dose decoctions and low dose decoctions compared significant difference(P<0.05).Conclusion:In study,traditional Chinese medicine compound in different analgesic decoction can significantly inhibit the EMs model of ectopic endometrium tissue pathologic hyperplasia in rats,the main mechanism lies in:A,traditional Chinese medicine compound in different analgesic soup can effectively lower the EMs model rats serum estrogen E2 and organization level of estrogen receptor,which block the estrogen receptor as signaling activation combined with estrogen and estrogen response element(ERE)combined with,Make the ectopic lesions cell growth and proliferation inhibition.Estrogen E2 levels 2,EMs model rats with cell membrane receptor,and make the Ras-Raf activated and downstream substrates ERK phosphorylation,phosphorylation of ERK after entering the nucleus to activate nuclear transcription factor nf-kappa Bp65,nf-kappa Bp50,c-Jun,c-the expression of Fos,Cause abnormal proliferation of cells,and compound within different analgesic soup can at the same time by EMs the ectopic lesions tissue in rat models of the nf-kappa Bp65,nf-kappa Bp50,c-Jun and c-Fos protein levels so that ectopic lesions cell growth and proliferation inhibition.Three,EMs model of ectopic endometrium tissue of rats significantly increased estrogen receptor levels,activating protein 1(AP-1)sub-Jun and c-c Fos,at the same time,the expression of aromatase P450 rise,cause ectopic lesions tissue cell proliferation,differentiation,angiogenesis,And traditional Chinese medicine compound within different analgesic soup could inhibit-Jun and c-c Fos gene transcription level and protein expression and inhibiting the activity of aromatase P450,thus improve the endometriosis ectopic lesions apoptosis rate,inhibit the excessive proliferation and control the development of endometriosis. |
