In basic science field of Biochemistry,industrial materials,medicine,it is more and more necessary to explore the microstructure of materials with optical microscopes which have these characteristics,higher resolution(up to micrometer,nanometer),large field,video speed,low cost,three-dimensional imaging,live cell imaging,label-free.Surrounding these issues,this paper focus on the platform of the reflectance confocal microscopy(RCM),stimulated Raman scattering microscope(SRS),Fluorescence imaging.The key researches are as follows.1.The structure and optical imaging principle of RCM system is studied.The4f structure,The optical magnification,optical resolution,scanning angle, imaging area and pinhole size are analyzed.The optical design process of ZEMAX is briefly described,at the same time,variable scale evaluation method is used to evaluate the whole optical structure performance in the theory exploration.2.The 2D scanning imaging mechanism of RCM is studied,and two schemes are realized by using the key components of high-frequency resonance galvanometer and polygon mirror.The advantages of the dual trigger mode (line signal + frame signal)of the data acquisition card and the timing synchronization relationship is described in detail.3.In order to obtain a large mosaic field,two stepper motors are used in the X-Y plane of RCM,and a high-precision stage is used in the X-Y plane of SRS.4.The high frequency noise from resonant galvanometer and laser heat dissipation are studied.5.The imaging platform of(SRS)is built.and the live cell imaging device of SRS was realized with commercial device modules and open source software.The development of RCM and SRS in this paper are of certain innovation and practical engineering value.
