PurposeTo explore the effect of Yiqi Huatan Huoxue Recipe on gastrointestinal mucosal injury and gastrointestinal motility decline induced by hypothyroidism in rats,and to elucidate the therapeutic effect of Yiqi Huatan Huoxue Decoction on prevention and treatment of gastrointestinal mucosal injury and gastrointestinal motility.Provide new ideas for clinical prevention and treatment of hypothyroidism and related symptoms.Materials and Methods1.Theoretical researchOrganizing and arranging the etiology,pathogenesis and treatment of related diseases such as "cracking","edema","virtual labor" and "five late" from the pre-Qin and Han dynasties to the Ming and Qing Dynasties,and searching for hypothyroidism in modern literature.The prevention and treatment of the etiology and pathogenesis of the disease,explain the theoretical research on the hypothyroidism.2.Experimental StudyEighty SPF Wistar male rats were established by PTU gavage to establish a model of hypothyroidism in rats.The experimental rats were randomly divided into 7 groups: normal group(8),model group(12),and LT-4 group(12),LT4+ Chinese medicine high-dose group(12),LT-4+ Chinese medicine low-dose group(12),Chinese medicine high-dose group(12),and traditional Chinese medicine low-dose group(12).Taking Yiqi Huatan Huoxue Recipe as treatment drug,LT-4 was used as control drug.Each group was fed for 8 weeks according to the corresponding dose,taking serum,stomach tissue and pylorus to rectal end and colon tissue.The expression of T4 and TSH in rat serum was determined by radioimmunoassay.The expressions of TFF2,TFF3,EGF and GLP-1 protein in colonic tissues of rat gastric antrum were determined by ELISA.The expression of GLP-1 mRNA in colon tissues of TFF2,TFF3 and EGF in gastric antrum of each group was determined by qRT-PCR.The gastric emptying rate and the small intestine propulsion rate were measured by activated carbon gavage method.The data were statistically analyzed using SPSS-17.0.Result1.Theoretical researchThe hypothyroidism of traditional Chinese medicine belongs to the categories of"sickness","edema","Fiction" and "Fullness"."Constipation" The pathogenesis and pathogenesis are related to four aspects: kidney deficiency,spleen and stomach loss,liver qi deficiency and blood stasis.Treatment should be based on the cause of the disease,syndrome differentiation and treatment,taking into account the specimens,using the methods of invigorating the kidney and strengthening the spleen,nourishing the liver and nourishing blood,promoting blood circulation and removing blood stasis,and exerting the advantages of TCM as a whole and dialectical treatment.Our research team used Yiqi Huayu Huoxue Recipe to treat the hypothyroidism theory.2.Experimental Study2.1 Radioimmunoassay for serum T4 levels before and after treatment in each group of ratsBefore treatment: Compared with the normal group,the serum T4 levels in the model group and each treatment group were significantly decreased(P<0.01),and there was no significant difference between the treatment groups(P>0.05).After treatment: compared with the normal group,the serum T4 content of the model group and each treatment group were significantly different(LT4+ high dose group of traditional Chinese medicine P<0.05,P<0.01);Compared with the model group,the expression of serum T4 in each treatment group was significantly up-regulated(LT4 group,high-dose Chinese medicine group,low-dose Chinese medicine group P<0.05,P<0.01);The comparison of the drug groups showed that the serum T4 expression was slightly lower in the low-dose group(Ps LT4+ Chinese high-dose group P<0.01),and there was no significant difference between the other groups(P>0.05).2.2Radioimmunoassay for serum TSH levels before and after treatment in each group of ratsBefore treatment,serum TSH levels were significantly up-regulated in the model group and each treatment group compared with the normal group(P<0.01).After treatment,compared with the normal group,the expression of TSH in the model group and each treatment group was significantly different(P<0.01);Compared with the model group,theserum TSH expression of each drug treatment group was significantly down-regulated(P<0.01);The comparison of the drug groups showed that the high-dose group of LT4+Chinese medicine had the best trend of down-regulating TSH expression.2.3Comparison of mRNA expression of TFF2,TFF3 and EGF in each groupCompared with the normal group,the expression of TFF2 mRNA in the model group and each drug treatment group were significantly different(P<0.01);Compared with the model group,except for the low-dose group of traditional Chinese medicine(P>0.05),the TFF2 mRNA of the other drug treatment groups were significantly up-regulated(P<0.01);Comparison of the drug groups showed that the high dose of LT4+ Chinese medicine had the best effect on up-regulating TFF2 mRNA.Compared with the normal group,the expression of TFF3 mRNA in the model group and each drug treatment group were significantly different(P<0.01).Compared with the model group,TFF3 mRNA was significantly up-regulated in each drug treatment group(P<0.01).Comparison of the drug groups showed that the high dose of LT4+ Chinese medicine had the best effect on up-regulating TFF3 mRNA.Compared with the normal group,the expression of EGF mRNA in the model group and each drug treatment group were significantly different(P<0.01).Compared with the model group,EGF expression was significantly down-regulated in each drug treatment group(P<0.01);Comparison of the drug groups showed that the low dose of LT4+ Chinese medicine had the best effect on down-regulating EGF mRNA.2.4Comparison of the expression of TFF2,TFF3 and EGF protein in each groupCompared with the normal group,the expression of TFF2 in the model group and each drug treatment group were significantly different(P<0.01);Compared with the model group,except for the low-dose group of traditional Chinese medicine(P<0.05),the TFF2 of the other drug treatment groups were significantly up-regulated(P<0.01);Comparison of the drug groups showed that the high dose of LT4+ Chinese medicine had the best effect on up-regulating TFF2.Compared with the normal group,the expression of TFF3 in the model group and eachdrug treatment group were significantly different(P<0.01);Compared with the model group,each drug group(P<0.01);compared with the LT4 group,except the LT4+ high-dose group(P<0.05);Compared with the high-dose group of traditional Chinese medicine,except for the low-dose group of LT4+ Chinese medicine(P<0.05),the TFF3 of the other drug treatment groups were significantly up-regulated(P<0.01).Compared with the normal group,the expression of EGF in the model group and each drug treatment group were significantly different(P<0.01);compared with the model group,each drug treatment group(P<0.01);EGF was significantly up-regulated in the other drug treatment groups(P<0.01);The comparison of the drug groups showed that the high dose of LT4+ Chinese medicine had the best effect on up-regulating EGF.2.5Differential expression of GLP-1 mRNA and protein in colon tissue of each groupCompared with the normal group,the expression of GLP-1 mRNA in colon tissue was significantly increased(P<0.01).Compared with the model group,the activity of GLP-1protein in the colon of each drug treatment group was significantly down-regulated(P<0.01);Comparison of the drug groups showed that the high dose of LT4+ Chinese medicine had the best effect on down-regulating GLP-1.Compared with the normal group,the expression of GLP-1 protein in colon tissue was significantly increased(P<0.01).Compared with the model group,the activity of GLP-1protein in the colon of each drug treatment group was significantly down-regulated(P<0.01);Comparison of the drug groups showed that the high dose of LT4+ Chinese medicine had the best effect on down-regulating GLP-1.2.6Comparison of gastric emptying and small intestine propulsion rate in rats with hypothyroidismCompared with the normal group,the small intestine propulsion rate of the model group and the rats in each treatment group decreased significantly(P<0.01);Compared with the model group,the small intestine propulsion rate of the rats in each drug group was significantly increased(P<0.01);The comparison of the drug-treated groups showed that the small intestine propulsion rate of the rats in the high-dose LT4+ Chinese medicine group wasthe most obvious.Compared with the normal group,the gastric emptying rate of the model group and the rats in each treatment group decreased significantly(P<0.01);Compared with the model group,the gastric emptying rate of the rats in each drug group was significantly increased(P<0.01);Compared with the drug-treated groups,the gastric emptying rate of the rats in the high-dose LT4+ Chinese medicine group was the most obvious.Conclusion1.Hypothyroidism belongs to the category of "ricky disease","edema","Fiction","fullness" and "constipation".The occurrence of this disease is mainly related to spleen and kidney yang deficiency,liver stagnation and blood stasis and water drink internal stop.We believe that the most fundamental pathogenesis of hypothyroidism is "spleen deficiency and phlegm and poisoning",and there is a solid theoretical basis for the treatment of qi and phlegm and blood stasis.Therefore,under the guidance of the clinical experience of the instructor,this study combined with literature research to establish Yiqi Huatan Huoxue Decoction for prevention and treatment.2.Yiqi Huatan Huoxue Decoction can significantly improve the thyroid function of hypothyroidism rats,and gradually restore thyroid hormone levels to normal.3.Yiqi Huatan Huoxue Recipe protects the gastric mucosa of hypothyroidism rats by up-regulating TFF2 and TFF3 in gastric antrum tissue of rats with hypothyroidism and down-regulating EGF expression.4.Yiqi Huatan Huoxue Recipe can down-regulate the expression of GLP-1 in the colon tissue of rats with hypothyroidism,and increase the gastric emptying rate and intestinal propulsion rate.
