Purpose:By preparing a rat model of hypothyroidism,to investigate the mechanism of hypothyroidism leading to hepatic steatosis,and observe the effect of Yiqi Huatan Huoxue Decoction on non-alcoholic fatty liver caused by hypothyroidism.Material and method:1.Establishment of model: 80 SPF Wistar rats were adapted for one week,Eight rats were randomly selected as the normal group,and the remaining 72 were given intragastrically with a dose of 10 mg/(kg·d)of propylthiouracil(PTU),Four weeks after gavage model validation.2.Experimental grouping: After successful establishment of the model,72 rats were randomly divided into 6 groups(12 in each group): model group,traditional Chinese medicine constant group,traditional Chinese medicine high dose group,L-T4 group,and traditional Chinese medicine constant + L-T4 Group,traditional Chinese medicine high dose group + L-T4 group.3.Drug intervention: normal group fed with double distilled water;The model group and other groups given intragastrically with a dose of 10 mg/(kg·d)of propylthiouracil(PTU);The traditional Chinese medicine constant group was given to Yiqi Huatan Huoxue Decoction 13.7g/(kg·d)gavage;The traditional Chinese medicine high dose group was administered with Yiqi Huatan Huoxue Decoction Concentrate 41.1g/(kg·d)gavage;The L-T4 group was administered with left thyroid sodium suspension 5ug/(kg·d);The traditional Chinese medicine constant + L-T4 group was given 5ug /(kg · d)of suspension of left thyroid sodium tablets first,and then one hour later,13.7g /(kg · d);The traditional Chinese medicine dose + L-T4 group first left thyroid sodium tablet suspension 5ug/(kg·d),and then Yiqi Huatan Huoxue Decoction concentrated agent 41.1g/(kg·d);Every irrigation 6 days,rest day,for 8 weeks.4.Specimen collection and index detection: At the end of the 4th week,blood was collected from the rats' axillary veins,serum thyroxine(TT4)levels were measured by radioimmunoassay(RIA),and serum thyroid stimulating hormone(TSH)levels were measured by immunoradiometric assay(IRMA).At the end of the 12 th week,anesthetized rats were bled from the abdominal aorta,fresh liver livers were weighed to calculate the hepatic index,serum total thyroxine(TT4)levels were measured by radioimmunoassay(RIA),and serum by immunoradiometric assay(IRMA).Thyroid stimulating hormone(TSH)levels;Automatic biochemical analyzer detects triglyceride(TG),total cholesterol(TC),low density lipoprotein(LDL-C),high density lipoprotein(HDL-C),alanine aminotransferase(ALT),and Aspartate aminotransferase(AST)and liver tissue TG,TC levels;The rat liver was stained with oil red O and the morphology of hepatocytes was observed under light microscope.Results:1.After 4 weeks of model preparation,compared with the normal group,the level of TT4 in the model preparation group was significantly lower(P<0.01),and the level of TSH was significantly higher(P<0.01),suggesting that the hypothyroidism model was established;After 8 weeks of drug intervention,compared with the model group,serum TT4 levels were increased in each group of traditional Chinese medicine constant group,traditional Chinese medicine high-dose group,L-T4 group,traditional Chinese medicine constant + L-T4 group,traditional Chinese medicine high-dose + L-T4 group(P <0.05 or P<0.01),TSH levels decreased(P<0.05 or P<0.01);Compared with L-T4 group,serum TT4 level in high dose group was increased(P<0.05),and TSH level was decreased(P<0.05).2.Compared with the normal group,the levels of serum TG,TC and LDL-C in the model group were increased(P<0.05 or P<0.01);Compared with the model group,traditional Chinese medicine constant group,traditional Chinese medicine high-dose group,L-T4 group,traditional Chinese medicine constant + L-T4 group,traditional Chinese medicine high dose + L-T4 group serum TG,TC,LDL-C levels significantly decreased(P< 0.01);The levels of TG and TC in the Chinese medicine high dose group were significantly lower than those in the L-T4 group and the Chinese medicine constant group,and the difference was statistically significant(P<0.05).There was no significant difference in serum HDL-C among the groups,and the difference was not statistically significant(P>0.05).3.Liver power levels were higher in the model group than in the normal group(P<0.05);ALT and AST levels were higher in the Chinese medicine groups,L-T4 group,and the L-T4 group than in the model group.The difference was statistically significant(P<0.05 or P<0.01).4.Liver wet weight,liver index,Compared with the normal group,the hepatic wet weight and hepatic index were all increased in the model group(P<0.05).Compared with the model group,L-T4 group,each dose group of traditional Chinese medicine,L-T4+ traditional Chinese medicine group had lower liver weight and hepatic index(P<0.05 or P<0.01).There was no significant difference between the T4 group and the traditional Chinese medicine group.Compared with the L-T4 group,the liver index of the high traditional Chinese medicine group decreased significantly(P<0.05).In general,the best results were obtained with L-T4+ Chinese medicine high dose.5.Liver lipid levels were higher in the model group than in the normal group(P<0.05).L-T4 group,traditional Chinese medicine dose group,and traditional Chinese medicine + L-T4 dose group were decreased in different degrees compared with the model group(P <0.05 or P <0.01),traditional Chinese medicine high dose group was better than L-T4 group and constant group High level + L-T4 group has the best.6.Morphological observations,In the normal group,the edge of the liver cells was clear,the nucleus was intact,and no cytochrome-rich lipid droplets were seen in the cytoplasm-rich cytoplasm.but,in the model group,a large number of orange-red lipid droplets were scattered in the cytoplasm of hepatocytes,showing diffuse fatty changes.Compared with the normal group,the result of steatosis was statistically significant(P<0.01);Compared with the model group,the number of lipid droplets in the cytoplasm of hepatocytes in each dose group of traditional Chinese medicine,L-T4 group,each dose of traditional Chinese medicine + L-T4 group was decreased,and all had statistical significance(P<0.05 or P<0.01).Conclusion:Hypothyroidism can cause abnormalities in blood lipids,liver function,and fatty degeneration of the liver,resulting in the deposition of lipids;Yiqi Huatan Huoxue Decoction and L-T4 can improve the function of Thyroid function,correct lipid metabolism and reverse liver steatosis,and both drugs have synergistic effects.
