ObjectiveBy the analysis of serum bile acid metabolism spectrumof gallstones patients,in order to get serum bile acidmetabolites spectrum characteristics and metabolic markers may be involved in the molecular biology of network for different disease states,different TCM syndroem of gallstones patients,o explore the predictingendogenous metabolites of gallstones.MethodsClassifying 30 patients with gallstones with more than one year course as GS group,according to age and gender matched healthy 30 cases were classified as group H.Classifying 19 samples with of new onset gallstones patientsat the predisease point,as GS-0 group,according to age and gender matched healthy 19 cases were classified as group H-0.Classifying 19 samples with of new onset gallstones at the disease point,as GS-1 group.GS group and group H,GS-0 with H0 set of transverse comparison of bile acid metabolism spectral differences,and bioinformatics analysis.GS-1 with GS-0 group own longitudinal comparison of bile acid metabolism spectral differences,and bioinformatics analysis.Divideall the gallstone patients into liver Yin deficiency syndrome and liver qi depression symdrome by diagnostic criteria,after the match,compare two types of syndrome patients with gallstones and analysis the bile acid metabolism spectral differences by bioinformatics.Results1.Compared with healthy people,there are 10 differences in the bile acid spectrum of the GS groupgallstones patients.UDCAhas decreased,the level ofGCA,HDCA,TCA,GDCA,GCDCA,TCDCA,7-KetoDCAhave raised.Compared with healthy people,there are 12 differences in the bile acid spectrum of the GS-0 groupgallstones patients,the levelofGCA,HDCA,TCA,GDCA,GCDCA,HCA,TCDCA,7-KetoDCA,CA,CDCA,DCA,LCA have raised.Whether patients with gallstones or patients in the gall-stone predisease state,the serum bile acid metabolism of them are differenct with healthy people,the same converged and fluctuations of bile acid areGCA,HDCA,TCA,GDCA,GCDCA,TCDCA,7-KetoDCA.Compared with G1-0 group,the level of HDCA,7-KetoDCA,HCA in the GS-1 group have raised,the level of CDCA,LCA,CA,GHDCA,DCA,GDCA have decreased?Through the horizontal comparison,the GCA,HDCA have great contribution to the VIP value.Longitudinal comparison found that HDCA is the larger metabolic differences with high VIP contribution value,HDCA showed a trend of risein the disease states.2.the neutral bile acid metabolism pathway,FXR/RXR activation pathway,intrahepatic cholestasis involve the process,Biological molecular networks include: GATA3,GATA4,GATA6,GRB2,HNF4-alpha,KLF4,MYC,PAX1,POU5F1,SOX2,SOX7,TAF4,CEBPA,Sprr2,involving related diseases involving the bile acid synthesis dysfunction,abnormal weight loss,adenosine cyclophosphate synthetic anomaly,atherosclerosis.The upstream regulatory factors involvedGallstone new onset disease state to disease states include lactose and lipid.The pathology of biological changes and types includelean body mass,atherosclerosis,cholesterol metabolic.3.Gallstones liver Yin deficiency syndrome and liver qi depression syndrome,liver qi depression group of CA,LCA level rise,GHCA level down.Bile acid content involves the classical pathway of FXR/RXR spectral differences,PXR/RXR pathways,neutral bile acid metabolism pathway,intrahepatic cholestasis pathway,rBiological molecular networks include: GATA3,GATA4,GATA6,GRB2,HNF4-alpha,KLF4,MYC,PAX1,POU5F1,SOX2,SOX7,TAF4,CEBPA,Sprr2,and types of the biological changes intrahepatic cholestasis,bile acid synthesis disorder,cyclic amp synthesis,colon cancer cell activation,muscle fibroblast activation and DNA damage.Liver qi depression syndrome was more strongly with cholestasis.ConclusionHDCA may be able to predict bile acid to the pathogenesis of gallstone diseas in the process of the incidence of gallstones.Comparing the patients with Gallstones liver Yin deficiency syndrome and patientswith liver qi depression syndrome,the levelof CA,LCA level of liver qi depression group raise,GHCAlevel decrease.Metabonomics technology combination of bioinformatics technology is an effective research method of combining traditional Chinese and western medicine to explore complex pathogenesis.Provide the basis for early predict incidence of gallstones.
