Alkaloids are an important class of natural secondary metabolites.They are large in quantity,diverse in variety,rich in physiological pharmacological activity,and widely used in food,medicine and other fields.Although the fluorescent properties of certain alkaloids have been reported,the fluorescent properties of such compounds lacked in-depth systematic studies,and the fluorescent properties of many alkaloids have not been reported.In this thesis,the fluorescence properties of dozens of alkaloids were studied for the first time,on the basis of this,the relationship between the fluorescence properties and molecular structure of alkaloids was studied in depth,and new methods for fluorescence analysis of some active ingredients of traditional Chinese medicines were established.The research work of this thesis mainly includes the following three aspects:1.The fluorescence properties of more than 40 alkaloid compounds with different structure types were studied in detail for the first time,mainly investigated the influence of various environmental factors(include acidity or alkalinity of the solution,solvent polarity,ordered media,coexistent ions,placing and irradiation time,etc.)on on fluorescence performance.These alkaloids include: 15 simple apophine alkaloids from benzylictetrahydroisoquinoline alkaloids,such as glaucine,dicentrine,nantenine,nuciferine,aporheine,cassythicine,lauroscholtzine,xanthoplanine,boldine,norisoboldine,laurifoline,corydine,isocorydine,corytuberine and magnoflorine,et al;5 phenanthroine alkaloids,included sanguinarine,dihydrohydantraine,dihydroerythrazine,chelidonine and acetylcorynoline,et al;6 compounds from berberine and protoberberine alkaloids,which were tetrahydrocolumbamine,corypalmine,tetrahydroepiberberin,scoulerine,hydroprotopine and dehydrocheilanthifoline,et al;2 protopine compounds were protopine and allocryptopine;2 emetine alkaloids : emetine and cephaeline;and 4 other alkaloids derived from phenylalanine or tyrosine,such as lycobetaine,leonurine,higenamine and colchicine,et al;8 kinds of quinazolines and tructurally similar alkaloids,which included febrifugine,tryptanthrin,evodiamine,rutaecarpine,4-hydroxyquinazoline,2-methyl-4(3H)-quinazolinone,6,7-dimethoxy-4-quinazolone and quinazolone,et al.2.The fluorescence properties of more than 90 alkaloids studied in this paper and previously studied in this laboratory were summarized.These compounds are mainly coming from phenylalanine,tyrosine and o-aminobenzoic acids,covering almost all classes of endogenous fluorescent compounds in alkaloids.By comprehensively considering the influence of various environmental factors on the fluorescence spectra of these compounds,the relationship between fluorescence properties and molecular structure was analyzed and some regularity was obtained,which can be used to predict the fluorescence properties of some new alkaloid compounds.3.New fluorescence measurement methods were established for quantitative analysis of hydroxycamptothecine(HCPT)in HCPT injection and magnoflorine(MAG)in traditional Chinese medicine Qingfengteng.The reliability of the new methods was verified by HPLC or LC-MS/MS,and also showed that the fluorescence analysis methods are simple,accurate and reliable,which can provide new ideas for the analysis and determination of HCPT and MAG in traditional Chinese medicine or biochemical samples.
