| Chinese patent medicines(CPMs)is one kind of drug with different dosage forms,which is processed for the specified prescription and craft using Chinese Herb Medicines(CHMs)as raw materials with the guidance of traditional Chinese medicine(TCM)theory,and it has the characteristics of easy using,convenient storage and carrying.TCM fingerprints have the advantages of high sensitivity,good selectivity and strong fingerprint,which have gradually been applied to the identification and quality evaluation of TCM and proprietary Chinese medicines.Because of the good sensitivity,accuracy and selectivity,fluorescence spectrometry can be used to the qualitative and quantitative analysis of TCMs with fluorescent properties.The three-dimensional fluorescence spectra of CPMs for Jianpihewei,Qufengshi and Qingre were investigated in this thesis.The consistency of each kind of CPMs was studied by three-dimensional fluorescence spectra combined with cluster analysis and principal component analysis.Three-dimensional fluorescence fingerprints of three kinds of CPMs were established by average method,respectively.This dissertation consists of 5chapters.Chapter one:Introduction.The research progresses of three-dimensional fluorescence spectrum of TCMs etc.and the research status of three kinds of CPMs in recent years were summarized with 205 references.Chapter two:The optimization of experimental conditions.The experimental conditions including scan speed,EX/EM slit,PMT voltage and EX/EM sampling interval were optimized in this chapter and the optimum experimental conditions were of 2400 nm·min-1,5.0/5.0 nm,700 V and 5.0/5.0 nm,respectively.Chapter three:Study on the three-dimensional fluorescence spectrum of CPMs.The three-dimensional fluorescence spectra of 4 kinds of CPMs for Jianpi Hewei were determined,respectively.The consistency of the three-dimensional fluorescence spectra from each kind of medicine was investigated by cluster analysis and principal component analysis.The consistency was observed with a correlation coefficient of 0.905 for three-dimensional fluorescence spectra of 38 Fengliao Changweikang tablet samples from 3 manufactures.The consistency was obtained with a correlation coefficient of 0.856 for three-dimensional fluorescence spectra of 77 Weiyankang capsule samples from 3 manufactures.The three-dimensional fluorescence spectra of 40 Shugan Jieyu capsule samples from one manufacture gathered to the same category with a correlation coefficient of 1.000.The three-dimensional fluorescence spectra of 96 Qizao granule samples from 4 manufactures gathered to the same category with a correlation coefficient of 0.956.The results show that the three-dimensional fluorescence spectra of the four kinds of CPMs for Jianpi Hewei was in good agreement and the three-dimensional fluorescent fingerprints of the four kinds of CPMs for Jianpi Hewei were established by average method,respectively.Chapter four:Study on the three-dimensional fluorescence spectrum of CPMs.The three-dimensional fluorescence spectra of 4 kinds of CPMs for Qufengshi were determined,respectively.The consistency of the three-dimensional fluorescence spectra from each kind of medicine was investigated by cluster analysis and principal component analysis.The consistency was observed with a correlation coefficient of 0.984 for three-dimensional fluorescence spectra of 63 Fufang Fengshining table samples from one manufactures.The consistency was obtained with a correlation coefficient of 0.996 for three-dimensional fluorescence spectra of 40 Guli capsule samples from one manufactures.The three-dimensional fluorescence spectra of 63 Xuanqi Tongbi capsule samples from one manufacture gathered to the same category with a correlation coefficient of 0.997.The three-dimensional fluorescence spectra of 100 Tianma capsule samples from 7 manufactures gathered to the same category with a correlation coefficient of 0.880.The results show that the three-dimensional fluorescence spectra of the four kinds of CPMs for Qufengshi was in good agreement and the three-dimensional fluorescent fingerprints of the four kinds of CPMs for Qufengshi were established by average method,respectively.Chapter five:Study on the three-dimensional fluorescence spectrum of CPMs.The three-dimensional fluorescence spectra of 4 kinds of CPMs for Qingre were determined,respectively.The consistency of the three-dimensional fluorescence spectra from each kind of medicine was investigated by cluster analysis and principal component analysis.The consistency was observed with a correlation coefficient of 0.992 for three-dimensional fluorescence spectra of 72 Yinqiao Jiedu tablet samples from 3 manufactures.The consistency was obtained with a correlation coefficient of 0.993 for three-dimensional fluorescence spectra of 49 Siji Sanhuang tablet samples from one manufactures.The three-dimensional fluorescence spectra of 48 Qingre Tonglin capsule samples from one manufacture gathered to the same category with a correlation coefficient of 0.991.The three-dimensional fluorescence spectra of 91 Fukean tablet samples from 4 manufactures gathered to the same category with a correlation coefficient of 0.976.The results show that the three-dimensional fluorescence spectra of the four kinds of CPMs for Qingre was in good agreement and the three-dimensional fluorescent fingerprints of the four kinds of CPMs for Qingre were established by average method,respectively. |
