In alzheimer's disease microglia inflammation as the research object,adopting electroacupuncture "smell three stitches," observed the improvement on the alzheimer's disease model mice,"Smell three stitches" study on the alzheimer's mice learning memory improvement,the influence of MG cell inflammatory factor to restrain and the influence of signaling pathways,System of electroacupuncture "smell three stitches" biological mechanisms.Method:This paper is divided into two parts of the theoretical and experimental research.Theory research is partly through ancient and modern literatures on acupuncture and moxibustion treatment of AD and modern research progress review,analysis and mining,expected confirmed that acupuncture is an important treatment of AD.Experimental research part,first of all,using the APP/PS1 double transgenic mice,after adopting electroacupuncture "smell three stitches"intervention treatment in mice,Observe its behavior,MG cells in mice inflammation factors and the influence of signaling pathways,"smell three stitches" were discussed from molecular level to the mechanism of the AD,Discussion acupuncture stimulation on the biological mechanism of neurodegenerative diseases.Result:(1)the APP/PS1 double transgenic mice copied AD mouse model,model group rats learning and memory in cognitive situation,compared with the blank group,with significant difference.(P<0.05)(2)for AD mice,compared with the blank group,model group all can make their learning ability,after statistics processing,there is significant difference(P<0.01).After three needle and needle "smell" can obviously improve the learning and memory disorders,and has the characteristics of time dependent.(3)in hippocampus of mice MG cell inflammatory factor mRNA detection,after statistics processing,we found that the inflammatory cytokines IL-6,TNF-a,the expression of IL-1 beta,compared with normal control group,the AD model group mice inflammation factor expression significantly higher,with significant difference(P<0.01);Compared with the AD model group,intervention group "smell three stitches"inflammatory factor expression in mice significantly reduced(P<0.05);Compared with the AD model group,"smell three stitches + olfactory nerve to cut off the set of inflammatory factor expression did not change(P>0.05);Compared with the AD model group,donepezil HCL inflammatory cytokines expression of mice significantly reduced(P<0.05);Smell three stitches and donepezil HCL group was no significant difference(P>0.05).(4)in hippocampus of mice MG cell inflammatory factor of DNA testing,after statistics processing,we found that the inflammatory cytokines IL-6,TNF-a,the expression of IL-1 beta,compared with normal control group,the AD model group mice inflammation factor expression significantly higher,with significant difference(P<0.01);Compared with the AD model group,intervention group"smell three stitches" inflammatory factor expression in mice significantly reduced(P<0.05);Compared with the AD model group,"smell three stitches + olfactory nerve to cut off the set of"inflammatory factor expression did not change significantly(P>0.05);Compared with the AD model group,donepezil HCL inflammatory cytokines expression of mice significantly reduced(P<0.05);Smell three stitches and donepezil HCL group was no significant difference(P>0.05).(5)by Western blot test results show that compared with the blank group,a significant rise in AD model group mice hippocampus PI3K protein expression,with significant difference(P<0.01);Compared with the AD model group,intervention group "smell three stitches'hippocampus PI3K protein expression in mice significantly reduced(P<0.05);Compared with the AD model group,"smell three stitches + olfactory nerve to cut off the set of" hippocampus PI3K protein expression in mice did not change significantly(P>0.05);Compared with the AD model group,donepezil HCL group hippocampal PI3K protein expression in mice significantly reduced(P<0.05);Smell three stitches and donepezil HCL group was no significant difference(P>0.05).(6)by Western blot test results show that compared with the blank group,the AD model group mice hippocampus significantly increased AKT protein expression,with significant difference(P<0.01);Compared with the AD model group,intervention group "smell three stitches" hippocampus AKT protein expression in mice significantly reduced(P<0.05);Compared with the AD model group,"smell three stitches + olfactory nerve to cut off the set of"hippocampus AKT protein expression in mice did not change significantly(P>0.05);Compared with the AD model group,donepezil HCL group hippocampal AKT protein expression in mice significantly reduced(P<0.05);Smell three stitches and donepezil HCL group was no significant difference(P>0.05).Conclusion:(1)the APP/PS1 double transgenic mice can successfully replicate the AD model,the model has a good stability.(2)for AD model mice,and its cognitive memory data with time,the effect of degree of equal relationship between were positively correlated."Smell three stitches" intervention group compared with model group,the mice can significantly improve the cognitive ability of mice.(3)the experimental results show that MG in mice hippocampal inflammation factor beta,IL-6,IL-1 TNF alpha release of promote the occurrence and development of the AD."Smell three stitches" intervention group compared with model group,the mice could significantly inhibit the release of inflammatory cytokines.(4)for AD model rats,the hippocampus of PI3K and AKT protein expression and inflammatory factor expression,the effect of degree of equal relationship between positively correlated,and the "smell three stitches"intervention group mice can make the expression of PI3K and AKT protein close to normal.
