Objective: To reveal the relationship of electric acupuncture and excitatory amino acids receptor expression, and to investigate the mechanism of the influencing of electric acupuncture on learning and memory.Methods: The rats were randomly divided into the control group,the brine group,the model group,the medicine group and the electric acupuncture group; The method of preparation for animal model was injecting STZ into lateral cerebral ventricle."BAIHUI","DAZHUI","ZUSANLI","TAIXI"and"SHENSHU"were selected by electric acupuncture group for treatment. Detection index as follows: To evaluate the learning and memory ability of every group rats by means of Morris water maze; to observe the arrangement of neuronal cells of hippocampus by means of HE staining method; to detect the Aβprotein expression by means of immunohistochemistry; to detect the NMDA receptor subtypes NR1 and NR2BmRNA of hippocampus by means of fluorescent PCR.Results: 1,In position navigation test, the average latency of the model group rats was significantly increased than the control group, the electric acupuncture group was significantly reduced than the model group; in space exploration test, the number of times crossing platform of electric acupuncture group were significantly more than model group. 2,The number of neuronal cells of hippocampus reduced by means of HE staining method, pyramidal cells ordered irregularly,attenuated,abscised,decreased in the number, as well as inflammatory cells and so on. The number of neuronal cells of hippocampus were more than model group after the treatment of electric acupuncture, and cells ordered regularly, construction compact, morphology normal. 3,The Aβprotein expression of model group significantly increased, the expression was lower after the treatment of electric acupuncture. 4,After the treatment of electric acupuncture, the expressions of NMDA receptor subtypes NR1 and NR2BmRNA of hippocampus were more than model group.Conclusion:1,To copy successfully the model of STZ-AD rats. 2,The electric acupuncture could strengthen the learning and memory ability of AD rats. 3,The treatment of electric acupuncture could obviously degrade the Aβprotein expression of model rats. 4,Electric acupuncture encouraging the expressions of NMDA receptor subtypes NR1 and NR2BmRNA of hippocampus maybe is one of the mechanism which improves learning and memory ability of AD rats.
