Objective1.This study focused on the correlation between TCM syndromes and the concentra tion of monocyte chemotactic proteinl(Monocyte chemoattractant 1 protein,MCP-1),m acrophage migration inhibitory factor(Macrophage migration,inhibitory factor,MIF),mi 1k fat globule epidermal growth factor 8(milk fat globule EGF factor ??,MFG-E8)i n dyslipidemia patients.In order to find the relationship between the TCM syndrome a nd inflammatory factors,this study provide the foundation basis for the standardization of syndrome differentiation.2.On the basis of studying on the characteristics of blood lipid indexes,atherogenic index of plasma(atherogenic index of plasma,AIP),we analyzed the relationship between TCM syndromes and atherosclerosis(AS),also to provide a clinical basis for TCM treatment of dyslipemia and syndrome differentiation.3.On the research of metabolism modes in dyslipidemia patients to find the differences between TCM Syndromes,to obtain the metabolite,metabolic profile,metabolic pathway in different syndromes,futher to find the changes of Metabolic network,we want to rich the scientific connotation of syndrome and the system of patterns and treatment,to provide the basis for the development of syndrome by analyzing the correlation and variation between characteristic metabolites.It has important significance to provide a clinical basis for TCM treatment of dyslipemia.analysis the differences of metabolic mode between dyslipidemia and health,to find the metabolite,metabolic pathway in dyslipidemia.Methods1.Cross-sectional study design was used.120 patients with dysilipidemia were assigned with corresponding TCM syndrome scores and their MCP-1,MIF and MFG-E8 serum content measured with the method of Elisa.We also explored the characteristics of MCP-1,MIF and MFG-E8 in different TCM syndromes and their relativity with syndrome score in patients.2.Cross-sectional study design was used.120 patients with dysilipidemia were assigned with corresponding TCM syndrome scores and their Blood fats serum content measured in our hospital'clinical laboratory.We also explored the characteristics of Blood fats and AIP in different TCM syndromes and their relativity with syndrome score in patients.3.Cross-sectional study design was used.54 patients with dysilipidemia were assigned with corresponding TCM syndrome scores.10 healthy person were assigned at the same time.There are 18 case of liver kidney yin deficiency syndrome(GSYX),18 cases ofqi stagnation and blood stasis(QZXY),18cases of yin deficiency and yang hyperactivity(YXYK).After serum samples have been collected,metabonomics were detected in dyslipidemia patients and healthy person by using GC-MS,GC-TOFMS.To analsis multivariate data with SIMCA.We obtiain metabolic profiling by PCA,OPLS-DA,and then we draw load diagram.We get Differences of compound by KEGG.We also analsis hierarchical clustering,metabolic pathway and topology analysis of metabolic pathway with MetaboAnalyst 3.0.Results1.Inflammatory factors are correlated to different TCM syndromes:? Experimental group's concentration of inflammatory factors is higher than control group,P<0.05.?T he concentration of inflammatory factors is different in different TCM syndromes:MCP-1:YXYK>GSYX>QZXY;MIF:YXYK>GSYX>QZXY;MFG-8:QZXY>GSYX>YXYK?Th e scores of different TCM syndromes are related to inflammatory factors:a.The scores of GSYX is positively related to MCP-1(r=0.322,P=0.043),MFG-E8(r=0.322,P=0.04);b.The scores of YXYK is positively related to MCP-1(r=0.324,P=0.042),MFG-E8(r=0.321,P=0.043);c.The scores of QZXY is positively related to MIF(r=0.366,P=0.020).2.The study on the relationship of TCM syndrome with blood fat and AIP:?a.serum lipid parameter:The TC,TG,HDL,LDL levels of experiment group were higher than the control group,and there was statistical difference,P<0.05.b.The scores of TCM syndromes are related to serum lipid parameter:The scores of GSYX is positively related to TC(r=0.419),The scores of YXYK is negatively related to HDL(r=-0.413),The scores of QZXY is negatively related to HDL(r=-0.282)? the relationship of TCM syndrome with AIP:a.The concentration of AIP is different in different TCM syndromes:QZXY>GSYX>YXYK;b.The scores of TCM syndromes are related to AIP:The scores of GSYX(r=0.459,P=0.003),QZXY(r=0.358,P=0,023)is positively related AIP(r>0.3,P<0.05)?The occurrence of dyslipidemia is related to no stable occupation,age,BMI,family history.3.Differences of metabonomics features in TCM syndromes of dyslipimedia:?T he patterns of dyslipimedia and healhty person are distinct:a.Glycerol,1,3-diaminopropan,1,3-diaminopropan,2-Monoolein in dysilipimedia are higher than healthy person;b.The metabolic pathways are different in experiment group and control group:ABC transporte rs,beta-Alanine metabolism,Fatty acid biosynthesis.?The patterns are different in TCM syndromes of dysipimedia:a.There are 14 kinds of metabolites are different compared GSYX with healthy control group:stearic acid,Ribose,Glycerol,uric acid,phenylalanine,ci s-gondoic acid,2-Monoolein,lauric acid,3-Aminoisobutyric aci,3-Hexenedioic acid,1.3-dia minopropane,N-Acetylene-beta-alanine are up regulated;Monostearin,uridine are down r egulated;b.There are 33 kinds of metabolites are different compared QZXY with he althy control group:stearic acid,palmitic acid,linoleic acid,oleic acid,Glycerol,arachidonic acid,Hydroxylamine,uric acid,Cystine,heptadecanoic acid,lauric acid,1,2-Cyclohexanedion e,1.3-diaminopropane,Maleamate,N-Ethylglycine 2,N-Acetyl-beta-alanine are up regulated,while phenylalanine,2-Deoxytetronic acid2,cis-gondoic acid,Monostearin,2-Monoolein,d-G1 ucoheptose,Erythrose,Glucoheptonic acid,4-Hydroxybenzoic acid,3-Hydroxynorvaline 2,pi melic acid,beta-Glycerophosphoric acid,Hydroxyurea,Cyclohexylsulfamic acid,3-Phenyllact ic acid,3-ureidopropionate are reduced;Yin deficiency and yang hyperactivity group com pared with the healthy control group had 20 metabolites:Sixteen palmitic acid,oleic a cid,citric acid,glycerol,fourteen glucose-1-acid,phosphoric acid,lauric acid,trypto phan and other 1 kinds of material is raised,phenyl lactic acid,alcohol,phenylalanine,mustard salicylic acid and other 9 kinds of substances is reduced;c.There are 17kind s of metabolites are different compared YXYK with healthy control group:palmitic ac id,palmitoleic acid,citric acid,Glycerol,Myristic Acid,Glucose-l-phosphate,lauric acid,trypt ophan,1.2-Cyclohexanedione,1.3-diaminopropane2-Deoxy-D-galactose are up regulated,3-Phenyllactic acid,3-ureidopropionate,Sinapyl alcohol,phenylalanin,cis-gondoic acid,2-Mono olein,4-Hydroxybenzoic acid,3-Hydroxynorvaline 2,beta-Glycerophosphoric acid are dow n regluted.Conclusion1.The inflammatory factors of the abnormal blood lipid group were active than the normal group,we can combined with anti-inflammatory herbs in the treatment of abnormal blood lipid.The Objective indicators of QZXY are AIP,HDL;The Objective indicators of GSYX are AIP,TC,MCP-1,MFG-E8;The Objective indicators of YXYK are MCP-1,MFG-E8,HDL.2.Metabonomics studies show that patients with syndrome of dyslipidemia are dis tinguished,and screened the different metabolites and metabolic pathways.The patterns of experiment group and control group are different,we can try to use ABC transport ers,fatty acid biosynthesis,beta alanine metabolism pathway as recognition pathway in patients with dyslipidemia.Metabolic models are different in TCM syndromes of dysli pidemia,it proves the scientificity and distinguishability of TCM syndrome from the as pect of metabolism,and provides references for the objectification,visualization and m odernization of the TCM syndromes.
