There are two parts in the thesis.First part is theorizing the correlation between Diabetes cognitive disorder and sex hormone signaling pathway.Second part is scientific research:Kidney and Refreshing Method improves the diabetic cognitive disorder which is relates to regulate the function of "sex hormone signaling pathway".Theoritic explorationExplain the diabetic cognitive disorder's etiology, pathogenesis, syndrome differentiation and treatment, from the perspective of traditional Chinese medicine, tutor's views and my own research.Scientific researchObjective:To observe sex hormones'level of HPG, the level of Er? ? Er ? mRNAof type 2 diabetes mice's hippocampus. Thus clear the effect and mechanism of Kidney and Refreshing Method on improving cognitive impairment of type 2 diabetes mice's cognitive ability is related to its impact on the regulation of sex hormone signaling pathways in some links.Methods:8-week-old cleaning degree finale Kunming mice.They were given a basic diet as the normal control group.8-week-old SPF level finale KKay mice provided by the Chinese Academy of Medical Sciences Institute of Experimental. The OVX (ovariectomized)group was did ovariectomy.Then, all the KKay mice were given a high-sugar and high-fat diet.8 weeks later, the T2DM animal model was successfully set up. Then randomly divided them into model control group, positive control group, TCM(traditional Chinese medicine) group, except the OVX group. Positive group was given by gavage once a day with piracetam. And the TCM and the OVX groups were given by gavage once a day with Xingnaoyizhi agent. While all the KKay mice were given by gavage with the same volume of rosiglitazone daily to control the blood glucose. The gavage lasted for 56 days. We used Morris water maze to test the congnitive capacity of the mice before group and after drug. After the second water maze, do the experimental animal anatomy and sample preparation on the ice.The plasma levels of HPG hormone levels are detected by ELISA. Use QRT-PCR determination to detect the hippocampus's Er a and Er? mRNA. Then illuminate the correlation between HPG and "Pancreatic (spleen)-brain-kidney" axis's brain and kidney and the possible mechanism of Kidney and Refreshing Method is the regulation function of sex hormone signaling pathway. So to reveal the role of the Chinese medicine' targets and provide experimental basis for clinical treatment of the disease.Results:(1) The first time water maze experiment results:The search latency of normal control group and each group of diabetes mice was obviously shorten (P< 0.05).It is visible the T2DM cognition impairment mice cognitive ability significantly less than normal mice. The search latency of TCM OVX group and other groups of mice was obviously lengthen (P<0.05).It is visible the TCM OVX group cognitive ability significantly less than other groups of mice. The second time water maze experiment results:The search latency of normal control group was obviously shorter than model control and TCM OVX group (P<0.05). Comparing two times search latency:The second time water maze experiment results of TCM OVX group,positive control group and TCM group were much better than before (P<0.05). It illustrated both of piracetam and Xingnaoyizhi agent can improve the T2DM cognition impairment mice's cognitive ability. And Xingnaoyizhi agent can improve the TCM OVX group's cognitive ability.(2)In the study of HPG axis'hormones levels:Compared with the normal control group, the model control group's E2,P were obviously lower (P<0.05). It illustrated the T2DM cognition impairment mice's E2 and P were obviously lower than normal. The LH,FSH,PRL of normal control group and model control,positive control,TCM OVX group were obviously risen (P<0.05). And there is no significant change in T of every group.With Xingnaoyizhi agent treatment, the E2 and P of TCM group and TCM OVX group were obviously risen (The E2 and P of TCM and normal control group,TCM OVX and model control group were no obviously difference (P>0.05)). The LH and FSH of TCM group and TCM OVX group were obviously reduced (The LH and FSH of TCM and normal control group,TCM OVX and model control group were no obviously difference (P>0.05)). It illustrated Xingnaoyizhi agent can adjust HPG axis hormones disorder from improving E2 and P, and reducing LH and FSH.(3) In the study of the hippocampus's Er a and Er ? mRNA:Compared with the normal control group, the model control and TCM OVX group's Er? and Er?mRNA were obviously lower (P<0.05). The model control group's Er a and Er ? mRNA were obviously lower than TCM group's (P<0.05).There was no obviously difference between model control and TCM OVX group's Era and Er?m RNA (P>0.05). These illustrated Xingnaoyizhi agent can improve the hippocampus's Era and Er?m RNA of TCM and TCM OVX groups. Compared with the positive control group, the model control and TCM OVX group's Er a mRNA were obviously lower (P<0.05). It illustrated piracetam can improve the hippocampus's Era RNA of T2DM mice. There was no obviously difference between model control and positive control group's Er? m RNA(P>0.05). It illustrated piracetam had little influence on the hippocampus's Er ? RNA of T2DM mice.Conclusion:Based on conventional controling blood sugar, Xingnaoyizhi agents can improve the learning and memory capacity of the mice and the OVX mice with type 2 diabetic cognition impairment, with the possible mechanism of adjustment the disordered HPG:Adjusting the disordered levels of sex hormones, rising the levels of E2?P and reduced levels of LH?FSH?PRL.and then enhancing the Er a and Er ?'s mRNA expression in the hippocampus.
