Objective:Explore the function and mechanism of Buyang Huanwu Decoction on Caveolin-1gene knockout mice angiogenesis after cerebral ischemia.Methods:Focal cerebral ischemia mice model was duplicated by using middle cerebral artery occlusion.The species of experimental mice were mice with caveolin-1gene missing, homologous wild C57/BL/6mice,caveolin-1gene deficient mice which were named K,normal homologous wild C57/BL/6mice which were named N.Every species was divided into3groups, a total of6groups—sham operation group N,model group N,complementary yang group N,sham operation group K,model group K,complementary yang group K.Every group was given different treatment and neurological injury score of mice was elaluated on the3,7,14d; On day3,7,14mice were put to death, brain tissue cell morphology changes were observated by using HE dyeing methods, then MVD was calculated with VWF as markers. Receptor protein of VEGF and VEGFR-1, VEGFR-2and VEGFmRNA and receptor of VEGFR-1mRNA、VEGFR-2mRNA were detected by using immunohistochemical and qPCR. Results:(1)After cerebral ischemia,in two model groups the expression of VEGF and receptor of VEGFR-1and VEGFR-2were increased,which was lower in model group K,compared with model group N;(2)After cerebral ischemia,in two model groups the expression of MVD was increased which was lower in model group K,compared with model group N;(3)After cerebral ischemia, in model groups ischemia there were area structure disorder, irregular arrangement and cell swelling. Cell swelling was more apparent and new cells were less in model group K;(4)After cerebral ischemia,in two model groups the nerve function score were increased which was more obvious in model group K,compared with model group N;(5)Buyang Huanwu Decoction can significantly promote VEGF and the expression of receptor VEGFR1, VEGFR-2. compared with model group, the expression of knockout caveolin-1was weaker.(6)Buyang Huanwu Decoction can significantly promote the expression of MVD.Compared with model group, the expression of knockout caveolin-1was weaker. (7)Buyang Huanwu Decoction can promote brain cell populations.At14d,there were more new blood vessels in complementary yang group N compared with complementary yang group K.8)Buyang Huanwu Decoction can significantly enhance the recovery of function of model group. compared with model group, caveolin-1knockout resume was more slowly.Conclusion:(1)Caveolin-1knockout cells and nerve function injury model mice brain tissue is more serious.The microvessel density decreased, Buyang Huanwu Decoction can to some extent improve the above indexes.(2)Knockout of caveolin-1gene expression of mouse VEGF and its receptor in cerebral ischemic injury by.(3)Buyang Huanwu Decoctioncan promote neonatal ischemic vascularMay regulate caveolin-1/VEGF signal pathway.
