OBJECTIVE:The cerebral ischemia is the common disease of nerve system.Its pathology basic is that intumesce of ischemia brain tissues caused the intracranial pressure to increase which enhanced the ischemia and anoxyaemia,furthermore,evoke the brain cell to die.So improving the blood supply of ischemia zone is the key point.The vascular endothelial growth factor(VEGF) is the multifunctional agents effecting vascular endothelial cell,which participated in the recovery of ischemia brain tissues. BuYang HuanWu Decoction(BYHWD) is the classic prescription which protected ischemia brain injured and decreased mortality and mutilation rate through more path and more target.It investigated the effect of BYHWD on the cerebral ischemia through its intervention to new vessels,VEGF and its receptor of ischemia zone.METHODS:①Seventy five Sprague-Dawley rats(weight 280-300g) were randomly allocated to three groups:sham-operated group,model group and BYHWD group(drug group).There were 25 rats in every group. The cerebral ischemia rat model was made by occluding the middle cerebral artery(MCA) with thread.The BYHWD group was ingested with the liquid medicine of BYHWD after 2 hours of the operation(2ml/rat/day,according to the surface area,it was three times of the dosage of 70kg adult).All groups were executed on the 1d,3d,5d,7d,and 14d after the operation and once five rats.At the same time there were 3 rats as the normal control group.②The distribution and expression difference of VEGF and its receptor in the ischemia train tissues of rats in different groups have been investigated with immuohistochemistry technique.③The positive cells of VEGF were red in single labeling stain of immuohistochemistry technique. Four visual field was randomly selected on every brain slice in 10×10 or 40×10 field of microscope in order to counting the positive cells and getting the mean value.④The content of VEGF was detected with ELISA.The form of all data was mean±standard deviation and SPSS11.0 was used to analyse the measurement data(t test was used between two groups and ANOVA in multitude groups).RESULTS:①Expression of VEGF of hypoxia and ischemia brain tissues in BYHWD group was higher than those in the model group and sham-operated group.②The positive cells of VEGF in BYHWD group were obviously more than those in the model group on 7d and 14d(p<0.01).③Compared with model group,the expression of VEGF in BYHWD group was significantly stronger on 7d and14d(P<0.01).④The regularity of expression of VEGF in BYHWD group was the same with that in the model group.And except for 1d,it was higher at all the time points.Furthermore, Compared with model group,it was significantly different on 7d and 14d (P<0.01).CONCLUSIONS:The expression of the VEGF and VEGFR increased following ischemia injury in brain of rat,and the peak expression was observed on 3d.The postive cell was observed in the cortex and ischemia region.The morphous of postive cell is endotheliocyte and gliocyte. Compared with model group,the expression of VEGF and the amount of postive cell in BYHWD group were significantly different on 7d and 14d (P<0.01).BYHWD can enhance the expression of VEGF and promote the vascularization in brain ischemia,which giving satisfactory internal environment for nerve recovery and regeneration.BYHWD has therapeutical effect to cerebral ischemia.
