The Role Of Caveolin-1 In Inflammatory Response After Cerebral Ischemia And The Intervention Of Buyang Huanwu Decoction

Objective: To investigate the role of Caveolin1 in the inflammatory response after cerebral ischemia and effects of Buyang Huanwu decoction(BYHW)on expression of TNF-??ICAM-1 and IL-? after cerebral ischemia in Caveolin-1 knockout mice.Methods: 60Caveolin-1 knockout mice(KO)and 60 its cognate C57 BL / 6wild-type mice(WT)were randomly divided into three groups: sham group,model group and Buyang Huanwu Decoction group(20 mice in each group),and then randomly divided into four time points: Five animals were sacrificed at the four time points,respectively,at 3,7,10 and 14 days(5 at each time point).Using the middle cerebral artery occlusion(MCAO)in 2 hours to make the model of cerebral ischemia in mouse.The expressions of TNF-? ? ICAM-1and IL-? were measured by immunohistochemical staining.Results: 1.The contents of ICAM-1,TNF-? and IL-1? in Caveolin-1 knockout mice and C57 BL / 6 wild type mice in the model grou p were significantly higher than those in sham operation group(P < 0.01);2.The levels of ICAM-1,TNF-?,and IL-1? in the model group of Caveolin-1 knockout mice were significantly higher than those of C57 BL / 6 wild type(WT)mice model(P <0.05);3.The expression of ICAM-1 protein,TNF-? protein and IL-1? protein in Caveolin-1 gene knockout mice and C57 BL / 6 wild type mice in the Buyang Huanwu Decoction group were significantly lower than those in the model group at the same time point(P <0.05).Conclusion: 1.Caveolin-1 knockout can enhance the secondary inflammatory response of ischemic brain injury;2.Buyang Huanwu Decoction can regulate the expression of ICAM-1,TNF-? and IL-1? protein by Caveolin-1,so as to protect cerebral ischemia.
