Long non-coding RNA (lncRNA) is a kind of RNA which range from200bps to several thousand kb in size, transcribed by RNA polymerase II but don’t encode protein. They involve in all kinds of cellular activities. Till now dozens of lncRNAs have been related to tumors. In1999scientists found PCA3, a lncRNA that was specifically expressed in malignant tumors of prostate. Thus it has been used as a tumor mark for prostate cancer. However the function and mechanism of PCA3remain unknown. The aim of this study is to find out the function of PCA3by exogenously over-expressing PCA3RNA in PC3, a prostate cancer cell line, through transient transfection. After been transfected by pcDNA3.1(+)-PCA3plasmid, the proliferation rate of PC3cells accelerated (P<0.05), the migration was promoted (P<0.05) and the apoptosis rate declined (P<0.05). Through real-time PCR, we confirmed that the expression of PCA3RNA was elevated by thousands of time in PC3after been transfected by pcDNA3.1(+)-PCA3plasmid. Thus we come to a conclusion that PCA3RNA can promote cell proliferation, migration and also inhibit apoptosis.
