Theoretical Discussion On Treating AD From Lung And Experimental Research In Treating AD With BuFeiXuanFeiTongFuJiangZhuo Prescription

This thesis have carried out related theoretical discussion and preliminary experimental research in treating Alzheimers disease from Lung with TCM theory firstly at home. It also enriched the theory of etiology and pathogenesis,perfected the framework of treatment that prevent and treat Alzheimers disease with TCM, therefore offer a new thread and method for preventing and treating Alzheimers disease with TCM.In the part of theoretical research,we discussed the relations between lung and AD from two aspects. One aspect is from physiological function of the lung,the other is from the relations between lung and four other visceras. And based on the literature and clinical research material, we drew a conclusion that the deficiency and functional disorder of the lung play an important role in the happening and developing processes of Alzheimers disease. And we put forward a innovative viewpoint which is treating Alzheimers disease from lung and taking BuFeiXuanFei Tong FuJiangZhuo as the principle and method for treating AD.In the part of experimental research, established AD animal models,72 male SD rats were randomly divided into six groups with 12 cases in each group, namely, the normal group, the model group, the sham-operated group, the western medicine control group(Huperzine A group), BuFei XuanFeiTongFuJiangZhuo prescription group of low dose, and the BuFei XuanFeiTongFuJiangZhuo prescription group of high dose. After 28 days treatment, the rats were killed so as to detect the change of ethology by Morris water maze,the activity of AchE by chemistry chromatometry, the expression of pCREB by immunohistochemical method, and the cellular apoptosis by FCM method. The Objective of experimental research is to explore the therapeutic effect and acting mechanism of BuFeiXuanFei TongFuJiangZhuo prescription so as to verify the essentiality of treating AD from lung. The results signify that BuFeiXuanFeiTongFuJiangZhuo prescription can remarkably improve disturbance of ethology of the AD model rats, step down the activity of AchE in brain tissue, boost the ability to learning and memory,enhance the expression of pCREB and inhibit cellular apoptosis. Accordingly it suggest that the therapeutic effect of BuFeiXuanFei TongFuJiangZhuo prescription probably lies in protecting cholinergic system,improving the disorde of signal transduction of central nervous system and inhibiting nerve cells from apoptosis. The result also suggest that BuFeiXuanFeiTongFuJiangZhuo prescription is superior to Huperzine A and exists a certain dose-dependent relationship.By means of theoretical and experimental research,we drew a conclusion that the deficiency and functional disorder of the lung plays an important role in the happening and developing processes in Alzheimers disease. It is essential and feasible to treat AD from lung.
