Objectives:Studying the Bazheng San action mechanisms of the pathological model of β3-AR, Ki-67, p53,IL-6,IL-17,TGF-β1 of SD rats suffering from glandular cystitis,affected by escherichia coli, to provide scientific basis for the method of curing glandular cystitis by Bazheng San.Methods:Intervening the pathological model of Bazheng San used on SD Rat glandular cystitis,observing the common changes of the rats, testing the indexes of cystitis pathology and immunity, etc., to investigate the action mechanism of curing glandular cystitis by Bazheng San.Results:1. Compared with blank control group, the food intake and body weight of the SD rats in the other groups declined (P<0.05) Compared with blank control group and Bazheng San-duct group, the food & water intake and body weight of the SD rats in model group decreased obviously (P<0.01). Compared with HCPT group, the food intake of the SD rats in Bazheng San-duct low-dose group decreased obviously with statistical significance (P<0.01).2. The mucosa, muscular layer and chorion of bladder were normal without any denatured, dead and inflammatory cells infiltrating and with even carmoisine and smooth mucosa tissue. The bladder triangular area and Lamina propria of the SD rats in model group showed amount of inflammatory cells infiltrating, interstitial edema and focal necrosis, and Brunn nest and metaplasia of glandular epithelium appeared in the Lamina propria. A little inflammatory cells infiltrating in the bladder lamina propria of the SD rats in HCPT group showed obviously decreased, compared with model group. The pathological changes of Bazheng San-duct high-dose group showed basically same to HCPT group. The pathological changes of Bazheng San-duct medium-dose group showed less serious than that of model group, but its inflammatory cells infiltrating and Brunn nest in laminae propria of bladder mucosa and metaplasia of glandular epithelium were more serious than that of Bazheng San-duct high-dose group. The pathological changes of Bazheng San-duct low-dose group were basically same to model group.3.Compared with blank control group, the content of IL-6, IL-17, TGF-β1 in the bladder tissue of SD rats in model group was increased obviously (P<0.01).Compared with model group, the content of IL-6, IL-17,TGF-β1 in the bladder tissues of SD rats in HCPT group and Bazheng San-duct high-dose group decreased obviously (P<0.01).the content of IL-6,TGF-β1 in the bladder tissue of SD rats in Bazheng San-duct medium-dose group decreased (P<0.05). The content of IL-6, IL-17,TGF-β1 in the bladder tissues of SD rats in Bazheng San-duct low-dose group had no obvious difference(P>0.05).4. Compared with blank control group, the content of β3-AR in the bladder tissues of CG rats in model group had no obvious difference(P>0.05).. Compared with model group, the content of β3-AR in the bladder tissues of CG rats in Bazheng San-duct groups had no obvious difference(P>0.05).5.Compared with blank control group, the content of Ki-67 in the bladder tissues of CG rats in model group increased obviously (P<0.01).Compared with model group, the content of Ki-67 in the bladder tissues of CG rats in drug group and Bazheng San-duct high-dose group deceased obviously (P<0.05).6.Compared with blank control group, the content of P53 in the bladder tissues of CG rats in model group increased (P<0.05)Conclusions:1.The glandular cystitis of female SD rats will change pathologically by ducting Escherichia coli into bladder. The establishment of the pathological model helps to clinical, diagnosis and drug treatment of glandular cystitis.2. Bazheng San helps to reduce the content of IL-6, IL-17, TGF-β1. The content reduction of IL-6, IL-17 and TGF-β1 lies on the dosage of Bazheng San.3. The content of Ki-67 protein can be reduced and cell proliferation activity can be inhibited to a certain extent by Bazheng San.4. The content of β3-AR in the bladder tissues can not be reduced by Bazheng San in the experiment. The content reduction of β3-AR lies on dosage of Bazheng San.5. The content of p53 in the bladder tissues can not be reduced by Bazheng San. The prescription is not effective to the reduction of p53. The content reduction of p53 doesn’t lie on dosage of Bazheng San.
