Objective1、 acute bacterial cystitis mouse model, and use the detection index, verify whetherthe model has a good stability.2、observe the effect of sodium hyaluronate on bacterial cystitis tissue damage repaireffect, therapeutic effect of the optimum perfusion times.Methods1、 female7-9weeks of age,36Kunming mice, were randomly divided intoexperimental group and control group, with18rats in each group, two groups of micewith transurethral resection of bladder were perfusion50ul highly pathogenicEscherichia coli ATCC35218suspension of1-2*108cfu or50μ L PBS solution, afterperfusion6h,24h,72h, each time to collect the experimental group and the controlgroup with6rats in each group mouse bladder tissue, tissue homogenate wereobserved bacterial colony culture of bacteria; paraffin-embedded bladder tissuesections stained with HE, ordinary optical microscopy bladder tissue morphologicalchanges and bladder epithelial cell colony (IBCs) and inflammatory cells infiltration; transmission electron microscope bladder tissue intracellular bacterial colony (IBCs);to observe the morphological changes of bladder tissue and intracellular bacterialcolony organization immunofluorescence staining (IBCs). Model making8days all the mice and bladder tissues wereobserved by HE staining of bladder tissue fluorescence microscope ordinary opticalmicroscopy bladder tissue, WGC conjugated fluorescent antibody and nuclear dyes.Bladder tissue homogenate of inflammatory factor IL-5, KC determination.Results1、bacteria after6h of bladder tissue homogenate colony number was1.8±0.45*108cfu\/g, morphological examination showed a large number of neutrophil infiltration,edema, urinary bladder epithelial small stripping, residual bladder epithelial cellsshowed a large amount of intracellular bacteria (IBCs). The control group was notdetected bacteria bacterial culture, pathological changes of morphologicalexamination no bladder tissue;Bacteria after24h of bladder tissue culture of bacteriawas2.6±0.51*108cfu\/g, morphological examination showed infiltration ofinflammatory cells, edema, urinary bladder epithelium of stripping, epithelial cellsshowed a large amount of intracellular bacteria (IBCs). The control group was notdetected bacteria bacterial culture, pathological changes of morphologicalexamination no bladder tissue;Bacterial72h reperfusion after bladder tissuehomogenate colony number is0.03±0.014*108cfu\/g, morphological examination showed inflammatory cells decreased, the tissue edema, epithelial damage area isslightly less, epithelial cells are still in the intracellular bacterial mass (IBCs). Thecontrol group was not detected bacteria bacterial culture, pathological changes ofmorphological examination no bladder tissue.2、 perfusion of sodium hyaluronate1times or2times or3times after the mousebladder tissue compared with the positive control group, the inflammatory reactiondecreased, edema, urinary bladder epithelial damage, loss was higher than the positivecontrol group. Perfusion with2times the damage degree of bladder tissue wassignificantly lower than that of reperfusion1, inflammatory factor IL-5, KC levelswere significantly lower than the perfusion of1times, there are statisticallysignificant, P<0.05. Infusion of HA2and3mice, the negative control group micebladder tissues showed no significant injury, inflammatory factor level had nosignificant difference.Conclusion1、7-9weeks of age, female Kunming mice transurethral perfusion of highlypathogenic Escherichia coli ATCC3521850ul1-2*108cfu can be successfullyproduced acute infection mice model.2、 sodium hyaluronate, obvious tissue repair effect on bacterial cystitis can reducethe degree of inflammation of bladder tissue.
