Objective: To observe the influence of electroacupuncture treatment on functional dyspepsia rats in general evaluation,gastrointestinal dynamic changes,small intestine driving rate,both central and peripheral related brain gut peptide:Ghrelin and Neurotensin expression.To investigate the role of brain gut peptide of ghrelin and neurotensin and explore the adjustment mechanism of electroacupuncture treatment on functional dyspepsia rats.Methods: 40 SPF Sprague-Dawley male rats(weight 200±20g)were randomly divided into four groups(n = 10 each): a normal group,a model group,a drug group and an electroacupuncture group.Except for the normal group,the other three groups underwent modeling totally 14 days by tail clamped stimulation(3 times/d),giving an irregular diet(feeding every other day,with free access to water),and gaving 0? distilled water.As FD model rats was successfully induced,Drugs and electroacupuncture treatment started respectively.Rats of Drug group were given diluted solution of Mosapride Citrate Tablets,Each dosage(10 ml/kg),once a day,six times a week,a total of 28 days for four weeks.Rats of electroacupuncture group were acupunctured at Zusanli(ST36),nerve stimulator mechine were conected with Zusanli,continuous wave,4 hz,2 m A were designed in the medicine,20 minutes once a time,with the same treatment course of drug group.After 28 days,the rats were killed to take tissue samples.The rates of gastric emptying and small intestinal transit were determined;general situations and changes of diet and drink of rats were observed and recorded,,Plasma content of ghrelin and NT were determinated by ELISA;expression levels of ghrelin and NT of each rat in gastric and hypothalamus were determinated respectively by Western blotting.Results:Observing the general situations and behavior of rats :Before modeling phaze,each group rats in the mental state and activity are good,fur shiny and neat,stool was dry globosity.In modeling phaze,model group,drug group and the electroacupuncture group rats fur withered knot,activity decreased significantly,arrest unresponsive,stool was watery yellow.Compared with the nomal group,the three other group rats in general score,body weight growth,growth of food per day were significantly decreased(P<0.05);After treatment phaze,drug group and electroacupuncture group rats fur to restore luster and tidy,good activity,food intake increased significantly,response for catching quick,stool turned to dry brown globosity,general score,growth of food per day and body weight growth were significantly increasing(P <0.05);There was no significant difference between two groups(P >0.05);2.Gastrointestinal dynamics change:(1)gastric residual rate: compared with normal group,model group rats of gastric residual rate increased significantly(P<0.05);Compared with model group,the gastric residual rate in electroacupuncture group was obviously lower(P<0.05),There was no significant difference between drugs,electroacupuncture group(P>0.05);(2)small intestinal transit rate: compared with normal group,model group rat of small intestine propulsion rate significantly decreased(P<0.05);Compared with model group,the electroacupuncture group of small intestine propulsion rate increased significantly(P<0.05),There was no significant difference between two groups(P>0.05);3.Effect of the plasma density of Ghrelin and NT : compared with normal group,model group rats plasma levels of ghrelin significantly decreased(P<0.05),the plasma levels of NT increased significantly(P<0.05);Compared with model group,plasma levels of ghrelin in drug group and electroacupuncture group rats increased significantly(P<0.05),the plasma levels of NT significantly decreased(P<0.05);Compared with the drug groups,the plasma levels of ghrelin in electroacupuncture group rats rise,the plasma levels of NT decreased,there was no statistically significant difference between two groups(P>0.05);4.Effect of protein expression of Ghrelin and NT in gastric and hypothalamus : compared with normal group,the expression of Ghrelin in gastric and hypothalamus in model group rats decreased obviously(P<0.05),the expression of NT in gastric and hypothalamus significantly increased(P<0.05);Compared with modelgroup,the expression of Ghrelin in gastric and hypothalamus in drug group and electroacupuncture group rats increased obviously(P<0.05),the expression of NT in gastric and hypothalamus in drug group and electroacupuncture group rats decreased obviously(P<0.05);Compared with drug group,the protein expression of ghrelin in gastric and hypothalamus in electroacupuncture group rats rised,the protein expression of ghrelin in gastric and hypothalamus decreased,but there was no statistically difference(P > 0.05).Conclusion: Our experiment successfully reproduced the closest to the clinical type FD rat model,according to symptoms and behavioral changes,suggesting the model established by the multifactorial intervention scheme was suitable for the diagnostic criteria of functional dyspepsia.With a significant reduction of the expression of Ghrelin and a increased expression of NT both in the peripheral and central in model group rats,suggestting that pathogenesis of FD is closely related to the relevant Brain gut peptide secretion disorder.Electroacupuncture treatment has a significant therapeutic effect on FD rats,can significantly improve the FD generally behavioral change,increase food intake and body weight,and effectively enhance the gastric emptying and small intestinal propulsion so as to improve the gastrointestinal disorders;The electroacupuncture treatment has a effect on the rise of plasma levels and expression of ghrelin both in the peripheral and central,and has a effect on the decrease of plasma levels and expression of NT both in the peripheral and central,,there was no statistically significant difference between drug group and electroacupuncture group.Therefore,we believe that the gastrointestinal dynamic disd-ords is the important element in FD.Brain gut peptides plays an important role in the regulation of digestive organ activities,and regulate the gastric emptying.Electroacupuncture treatment by raising Ghrelin and decreasing NT in plasma and peripheral and central protein expression levels,has effect on the adjustment of ghrelin and NT secretion disorder,improving FD symptoms,behav-ioral changes,gastrointestinal disorders,accelerating gastric emptying,promoting digestion,and so as to achieve the purpose of treatment on FD.
