Objective: Based on traditional Chinese medicine theory, to study the therapeutic effecton insomnia sleeps, and further clarify the mechanism of sleeps Decoction in the treatment ofinsomnia. The mechanism of the effect of Chinese medicine on the treatment of insomnia inhumans is not clear yet, in this study, the target of Chinese medicine act on the body wasresearched, through the observation on the effect of cytokine hypothalamus pathologicalchanges sleeps on insomnia rats and the expression of IL-1β, hormone PGD2, its synthaseL-PGDS and its receptor DPR, GABA in the central nervous system and its receptorGABAARa1subunit.Methods:1. Clean Wistar rats were intraperitoneal injected with PCPA, copy the model ofinsomnia rats. The successful model rats were randomly divided shumian decoction into low,model, high dosage group, positive drug control drug (zaorenanshen capsule group),and ablank control group, with18rats in each group. Observed the general state, Rightingexperimental differences in rats sleep latency; and total sleep time by pentobarbital sodium.2. Optical microscopy and transmission electron microscopy was used in observation ofultrastructure of the cerebral cortex, and changes in brain structure Shumian side of insomniain rats.3. The content of enzyme linked immunosorbent assay of IL-1β, hormone PGD2,neurotransmitter GABA;4. The expression was detected by fluorescence quantitative PCR PGD2mRNA,L-PGDS mRNA, DPR mRNA, GABAARa1m RNA.5. Using immunohistochemical method to observe the expression of neurotransmitterGABA.Results:1. Shumian decoction can improve the general state of insomnia in rats, reduce sleeplatency,and increase their total sleep time.2. Shumian decoction model with improved cerebral cortex yuan insomnia role of nervepathology.3. Shumian decoction increased the hypothalamus of rats cytokines IL-1β content. 4. Shumian decoction has the effect of improving insomnia expression in thehypothalamus of rats PGD2levels and PGD2mRNA, L-PGDS mRNA, DPR mRNA’s.5. Shumian decoction had the hypothalamic GABA content and GABAARa1mRNAexpression in rat models to improve the role of insomnia.Conclusion:Shumian decoction can improve the general state of insomnia in rats and reduced sleeplatency increased their total sleep time; Improve the structure of the cerebral cortex pathologymodel of insomnia. Insomnia can be improved while the hypothalamus of rats cytokinesIL-1β levels, insomnia hypothalamus of rats under PGD2levels and PGD2mRNA, theexpression of L-PGDS mRNA, DPR mRNA’s, Hypothalamic GABA levels and mRNAexpression in rat model GABAARa1improve insomnia. Description Shumian decoctionintervention insomnia plays a role in improving sleep through the hypothalamus of ratscytokines, hormones, neurotransmitters.Professor Wang believes the key to the pathogenesis of insomnia is lack of blood, uneasy.The essence of Chinese medicine is the overall concept.It’s to explore the whole rule of lifeactivities. In the body of the multi-system, multi-target therapy has its good overall place.Currently limited to a single multi-pharmacological studies on the role of Chinese medicinetreatment of insomnia. Chinese medicine can not fully reflect the overall regulationadvantages. And this experiment can study from multi-channel, multi-target, multi-facetedrole of a neuro-endocrine-immune network. So damaged body function to restore balance,play to improve sleep disorders, the role of normal biological rhythms, Achieve the aim ofimproving insomnia. Shumian decoction in treatment under the guidance of YiqiYangxue andNingxinAnshen, Intervention by cytokines, hormones, neurotransmitters play a role toimprove sleep.
