Purpose: With the aid of experimental insomnia animal model, give the evaluation ofShumian fang for the treatment of insomnia. By observing the Shumian fang and positivecontrol drug diazepam of insomnia rats in hypothalamic neurotransmitter serotonin (5-HT),dopamine (DA) content of regulation and expression change of5HT1a, DRD2receptor mRNA.To explore its mechanism of action and Provide scientific basis for traditional Chinesemedicine treatment of insomnia.Method:1.60wistar rats were randomly divided into6groups, Respectively is: theblank control group, model group, diazepam control, Shumian fang high dose group, Shumianfang dose group, Shumian fang low dose group. each group of10, in addition to the normalgroup, the rest of the group are PCPA abdominal cavity injection copy insomnia rat model.2.Using enzyme-linked immunoassays single amine neurotransmitter in thehypothalamus was detected in the content of5-HT, DA; Using QPCR (Real-time policyPCR) technology measure5-HT receptor5HT1ain the hypothalamus, DA, DRD2receptormRNA expression level.Result:1. Model group, Shumian fang in the high dose group, the Shumian fang in themiddle dose group, low dose group Shumian fang and stability with the blank groupcomparison on sleep latency and sleep duration are different(p<0.05).2.5-HT in insomnia rat hypothalamus, the content of DA and5-HT receptor5HT1a, DA, DRD2receptor mRNA expression both in the form of: Shumian fang low dosegroup compared with model group, no statistical significance.(p>0.05).Shumian fang highdose group and Shumian fang middle dose group compared with model group had significantdifference(p<0.05). Shumian fang high dose group compared with the stable group of nostatistical significance(p>0.05).Conclusion: PCPA cause insomnia model rats in the experiment of sodiumpentobarbital sleep which the sleep latency period extend and total sleep time shortened. Thismeans that the models successfully. Experimental results confirmed that Shumian fang, highdose group can improve insomnia model in rat hypothalamus5-HT and the content of5HT1areceptor cut down the content of DA and its DRD2receptor. Which can be thought of as Shumian fang treatment effect is good by adjusting the hypothalamus within5HT1a, DRD2receptor expression of5-HT, the change of the content of DA,then to have the effect oftreatment of insomnia.
