ObjectiveFever, pain is a common human diseases. But the common antipyretic analgesics with varying degrees of adverse reactions, such as gastrointestinal discomfort, liver kidney damage, so the less developed side effects of antipyretic, analgesic drug is the hot spots of researchers. This experiment studies the active ingredients of antipyretic and analgesic effects on Cyperus rotundus, provide the material basis for the antipyretic and analgesic drug development and data support.MethodsAccording to the preliminary study,we found that the alcoholic extract from Cyperus rotundus may have the role of antipyretic and analgesic.On the basis of this study, we carried out as follows to find the active ingredient of Cyperus rotundus on antipyretic and analgesic effects:1. Identification of Cyperus rotundus herbsThrough the first edition of Chinese Pharmacopoeia,2010, Cyperus rotundus was distinguished, respectively, from the character identification and physical and chemical identification.2. To confirm the effective fraction of alcoholic Extract from Cyperus rotundus on antipyretic and analgesic action.Alcoholic Extract from Cyperus rotundus was first extracted by different solvent. Then the hot-plate and writhing method in mouce, fever in rats induced by dry yeast were used to study the analgesic and antipyretic action of the different solvent extraction of alcoholic Extract from Cyperus rotundus. To confirm the effective fraction of alcoholic Extract from Cyperus rotundus.3. Purification of Effective parts and identification of the results The silica gel column chrormatography, gel column chromatography, thin layer preparation were uesd, supplemented by the TLC and HPLC analysis, to isolated and purified the effective parts which was confirm by the pharmacodynamic test. Then the compounds was for structural analysis.4. Pharmacodynamics test of the active ingredientFever in rabbits induced by Endotoxin, writhing method in mouce, thermal stimulation of mouce were used to filter out the active ingredients of Cyperus rotundus on antipyretic and analgesic effects.5. Extraction and separation of the active ingredientAccording to pharmacodynamics test of antipyretic and analgesic, the active ingredients was filter out. Then to establish the method of extraction and separation of the active ingredient by silica gel column chromatography.Results1. Compared with the first edition of Chinese Pharmacopoeia,2010, the herbs was distinguished as the Cyperus rotundus. L. rhizomes.2. By the, petroleum ether fraction reduced writhing response, prolonged latent phaseds of foot licking and inhibited the fervescence in febrile model rats.3. With the silica gel column chromatography, thin layer preparation, and other means, compounds1-4were isolated from Cyperus rotundus.4. According to antipyretic analgesic three animal models, found that Cyperus play the active ingredients of antipyretic and analgesic effects of α-Cyperone.5. Through the study of extraction and isolation on α-Cyperone, Cyperus rotundus was extracted by95%ethanol first, then extracted by petroleum ether. At the end,silica gel column chromatography, petroleum ether:ethyl acetate as eluant to be used to as gradient elution.ConcluusionIn this study, alcoholic Extract from Cyperus rotundus was extracted by a variety of solvent.Then combined with three animal models of antipyretic and analgesic ations, the petroleum ether layer was filtered out to have the antipyretic and analgesic effect. By silica gel column chromatography and other separation means, compound1-4was separated from the effective parts. Pharmacodynamics test proved that α-Cyperone isolated form the petroleum ether layer play as the active ingredients of antipyretic and analgesic effects. Through the separation and purified test, Cyperus rotundus was extracted by95%ethanol first, then extracted by petroleum ether. At the end,silica gel column chromatography, petroleum ether:ethyl acetate as eluant to be used to as gradient elution. α-Cyperone will be done.
