| Treatise on Febrile Diseases includes hundreds of classic formulas written by Zhang Zhongjing,a well-known doctor in Chinese history and Xiao Chaihu Decoction(XCD)is known as its representative.The original prescription is often used in Japan,the modifition of the prescription is often used in China.Zhou Aijun,chief physician of the Dongguan Hospital of Traditional Chinese Medicine in the Guangdong Province of China,took XCD as basic prescription and added Coptidis rhizoma,Atractylodis macrocephalae rhizome,Poria to compose the so-called Modified Xiao Chaihu Decoction(MXCD).MXCD has been used clinically to treat hundreds of gastric ulcer patients and its highly significant treatment effect is extremely welcomed.On the basis of clinical curative effect,this paper adopts a multidisciplinary approach to screen the effective part of MXCD,and to research the pharmacodynamics and action mechanism of an active ingredient group of MXCD and then the chemical compoition of the effective parts were analyzed.The results explain the efficacy material base of the prescription,provides the scientific basis for its clinical curative effect,and laid a certain foundation for its further development and innovation of TCM varieties.Under the guidance of TCM theory,as MXCD as a whole,and according to the category of chemical components contained in each herb of MXCD and based on the clinical observations of MXCD when treating gastric ulcers.We extracted and separated MXCD into volatile oil,alkaloid,phenolic acid,saponin,polysaccharide,and other substance fractions.We then carried out bioactivity screenings by using anti-H.pylori and pharmacology experiments and identified volatile oil,alkaloid,phenolic,and saponin as the effective substances of the prescription.In an in vivo rat and mouse model,gastric ulcers were induced by using pylorus ligation,followed by administration of acetic acid and absolute ethanol.Subsequently,the substance fractions on pharmacodynamics were studied in gastric ulcers and all showed significant pharmacological activity.The results show that the active ingredients group of MXCD can significantly reduce ulcer index,total acidity,gastric juice and pepsin activity,and can increase SOD activity,inhibit lipid peroxide MDA increased.And the result also showed that it can reflect the therpeutic effects of the original prescription,and it has a certain significance for the explanation of traditional Chinese medicine compound compatibility of synergy theory.Based on previous studies,we combined the active substance fraction of MXCD,and the acetic acid-induced gastric ulcer model were used to investigate the mechanism on anti-gastric ulcer.Enzyme-linked immunoadsordent assay(tumor necrosis factor-?(TNF-?),prostaglandin E2(PGE2)and epidermal growth factor(EGF)),Nitrate reductase assay(Nitric Oxide(NO)),western blotting assay(Bax,Bcl-2,cleaved-caspase-3 and cleaved-PARP(poly(ADP-Ribose)polymerase)),immunohistochemical analysis of HSP-70,p-AKT and PCNA were also used to evaluate the anti-inflammatory,anti-apoptosis and healing properties of the active ingredients group of MXCD.The results showed that several mechanisms are involved in the antiulcer activity of AIG,such as increase of PGE2,NO and EGF content and reduce the content of TNF-?.The upregulation of HSP-70,p-AKT and PCNA seem to be directly linked to the healing of the active ingredients group of MXCD.Bax,Bcl-2,cleaved-caspase-3 andcleaved-PARP are also play an important role in this cicatrisation.Therefore the results provide scientiific experimental basis for clinical curative effect.Using GC-MS and LC-MS chromatographic techniques to analyzes the chemical composition.84 compounds were isolated from the volatile oil and65 compounds were identified.4,27 and 23 compounds were identified from alkaloid,phenolic acid,saponin fractions respectively.According to the literature retrieval,many compounds with biological activity were identified in our research.it is significant to the interpretation of the material basis. |
