Through the early antiviral drugs intervention to guinea pigs new-onset of genital herpes model,that is to say the intervention was before the virus going into sacral ganglion, then we can observe influence of the early treatment on the skin lesions, the integral faded and the content of ganglion virus.That is mean whether intervention can lower lesions integral, get rid of the virus, or block the state into the latent or not.[Method] Clinical trials:Genital herpes patients of venereal outpatient from2006to2010were counted.Then this part of the patient’s clinical research features were studied, including general data, incentive, symptoms, signs, treatment method of tracking and follow-up in ordered to summarize genital herpes patient epidemiological characteristics in the tianjin area.Basic test part.The early antiviral drugs intervention to guinea pigs new-onset of genital herpes model and effect were studid. First genital herpes virus standard strains were cultured and subcultured, at the same time materials were drawed from patients with clinical genital herpes lesions for PCR detection.Positive was vaccinated in VERO cells training,and TCID50was determined. The guinea pig genital herpes new-onset of model was established by the poisonous force and stability of the strong clinical plant.Then the test group was divided into three groups:the first group: antiviral drugs was given after inoculation immediately lasting for10days;The second group:antiviral drugs was given after inoculation in three days lasting for10days.The third group:antiviral drugs was given after inoculation in seven days lasting for10days.And a positive control group was set up to vaccinate vagina by HSV standard strains and the negative control group was set up to vaccinate vagina by the virus maintain fluid. Antiviral drugs were individed to three kinds:acyclovir, valaciclovir,genciclovir.The indicators were observed after the models were established:(1)The occurrence, development and resolvement of skin lesions and change of lesions integral value.(2) After inoculation virus in30days the guinea pigs had been to be put to death and sacral ganglia organization had been to taked to do fluorescence quantitative PCR in order to measure virus copy Number.[Result] Clinical trials parts:The genital herpes incidence of these five years in Tianjin area was fluctuates in23-36cases per year. The number of men was more than women’s. Age distribution indicated that20～39which sexual activity was of the most. GH patients with high school culture was priority accounting for57.33%. Men of genital herpes annual incidence was volatile.The majority were30-,culture in the primary education, professional to workers, sex partner in hotels,primarily.There was no obvious difference between New-onset GH and recurrent GH on gender.The New-onset GH were most unmarried. The recurrent GH were the local tianjin, higher education background and the cadre in the majority.Basis test part:Early intervention to guinea pigs new-onset of genital herpes model in different time had different treatment effects. The influence of intervention in lesions integral and the lesions subsidise time were different on immediate intervention,3days,7days of intervention group. Lesion fade time of intervention immediately group and intervention3days group were shorter than intervention in seven days group and positive control group. And, in the morbidity initial stage skin lesions’s integral quantity of immediate intervention group was lower than the other three groups below. At the middle and late stage:skin lesions’s integral quantity of immediately intervention group and3days intervention group were less than seven days intervention group and positive control group. In addition, through testing the guinea pig genital herpes model sacral ganglia HSV-DNA content, we can see that there were no significant differences among different drugs and different intervention time.But no matter intervene with acyclovir, valaciclovir or genciclovir all can reduce virus content compared with controls.[Conclusion] It was the first summarization2006-2010in the genital herpes epidemiological material in Tianjin. The number of men was more than women.The majority patients were low cultural degree, and mostly for the premarital sexual contact. The majority men of genital herpes were30-,culture in the primary education, professional to workers, sex partner in hotels,primarily.There were no obvious difference between New-onset GH and recurrent GH on gender.The New-onset GH were most unmarried. Genital herpes still has not been effectively controlled.We should be based on the popular new changes appearrance, formulate corresponding prevention measures.It was the first study that the effects of three different antiviral drugs in different time on guinea pigs new-onset of genital herpes were different. The earlier interventions,the lesions subsidise time is shorter, the smaller the integral value lesions, sacral ganglia HSV-DNA content is lower. Three different kinds of antiviral drugs comparison, function to reduce the lesions integral value of acyclovir was weaker than valaciclovir and genciclovir. But it was no statistical significance.In sacral ganglia HSV-DNA content, the effects of the three drugs have no difference. |
