Considering the actual situation of Shandong Province, the operation and management mechanism of the barrier environment in line with national standards laboratory animal facilities were studied, through the testing of the barrier environment laboratory animal facilities, the development and implementation of the SOPs, the management of facility dynamic run, the identification of autoclave sterilization effect and other aspects of Shandong Center for Disease Control and Prevention. The relatively standard management system and the SOPs of the barrier environment laboratory animal facilities had been formed.To test the implementation effect of the SOPs and further improve animal management. The barrier environment facility of Shandong Center for Disease Control and Prevention was tracking and monitoring by Shandong Laboratory Animal Inspection and Testing Center that Shandong Laboratory Animal Management Committee commissioned through " the dynamic detection experiments of the laboratory animal facilities in the barrier environment","the testing of the autoclave effects of the laboratory animal facilities in the barrier environment","the comparing experiments of dropped bacterial counts of static and dynamic laboratory animal facilities in general environment and barrier environment","the monitoring experiment of the barrier environmental parameters of part of laboratory animal facilities in Jinan,"The SOPs applications were assessed.1.The dynamic detected experiments of the barrier environment laboratory animal facilityThe environmental facilities were dynamic detected quarterly according to national standard (GB14925-2001), a total4times one year. The results showed that facility run up to the national standard in the static condition. The comparison resulted that many indicators meet national standards of static operation and detected that many indicators were increased relative to the static test in a way except the dust particles count rising on dynamic run-time(temperature3.45℃, relative humidity17.15%, air flow rate0.02m/s, noise1.28db, the ammonia concentration10.25mg/square meter, the number of bacteria falling0.86bacteria/plate).2. The detected test of laboratory animal facility autoclaved of the barrier environmentIn accordance with the SOPs operation of related equipments. Feed, padding, drinking water and work clothes through the high-pressure sterilization; the cages by high pressure sterilization and aqueduct disinfectant disinfection. The purpose is to inspect the SOPs normalization and operability during the operation of laboratory animal facilities in the barrier environment, and to find a reasonable microbiology control measures, not only to achieve the national quality standards of the laboratory animals and the environment facilities, but to reduce operating costs and labor intensity.The results showed that the experimental animals drinking water was stored in a clean for4days after121℃30min autoclaved,0bacteria/plate. Feed were in barrier clean room for5days after132℃sterilized for7min to ensure no bacteria. Litter132℃sterilized for7min under vacuum and placed7days without sterile to achieve sterilization effect. Clothes were stored in the sterilization barrier clean storage room for7d at132℃sterilized for5min without no bacterial growing. The of infection of keeping boxes autoclaved or several commonly used disinfectant in the immersion was very good without being sterile, maintaining7d of the life cycle of disinfectant.3. The compared test of bacterial counts of laboratory animal facility in the static and dynamic state of general environment and barrier environment.In order to test and improve the SOPs of personnel, animals, goods out and in the facilities of barrier environment, Dropped bacterial count were detected under static and dynamic state of the general environment and barrier environmental facilities every six months. The results showed, better static than dynamic in general environment(static2.57±0.95; dynamically18.47±5.43); barrier environment far less common environment in the similar padding and the breeding density with high efficiency filter clogging(Static0; dynamic oo).It is great advantage in the barrier environment after replacement of high efficiency filter; dry bedding much better than keeping only considering the case of bacteria. Static better than dynamic in barrier environment; it can be maintained in a good under the static state in barrier environment; barriers with the self-purification capacity of high efficiency filter clogging and weakening, self-purification capacity can be recovered after the replacement of high efficiency filter; the dropped bacterial count can be reduced significantly in their dependent ventilation cage(IVC) of the barrier environment, and the working conditions of feeders can be greatly improved.Particularly noteworthy, the locker room that was between the barrier and the outside were strict. We actively investigated the reasons and keep scientific cleaning when the dropped bacteria have detected at the3d and4th in air shower, the dropped bacteria were detected in the3d,4th and5th of the corridor, it was found that door and door lock of the corridor leading to the rooms loosed affected the air tightness. There was a small crack between double-Fei multi pressure cooker and the barrier wall at the5th test dropped bacteria found in the clean preparation room. The buffer was connected with the barrier and outside the channel, at the3d,4th and5th dropped bacteria detected, self-examination found that seal and lock effected tightness, high efficiency air curtain might be invalid, after the replacement of the highly efficient filter, it was the Ocfu one dish at the6th.4. The application and evaluation of the SOPsThe Testing Center of Shandong Laboratory Animal in the Medicine Safety Evaluation Center of Shandong University School, the Laboratory Animal Center of Shandong Medical University and Shandong Provincial Center for Disease Control and Prevention had conducted a one-year " the management SOPs of laboratory animal facility in barrier environment" and " the detected experiment of the barrier environment laboratory animal facilities parameters of Jinan " trial run in2007. It included mainly temperature, relative humidity, air velocity (air changes), pressure gradient, noise and ammonia concentrations and was made the relevant comparison of the data and the combination of the above fully into account the experimental data to verify the standard operating norms and operating results, also found that code of practice needs be improved and the improvement in the study had the desired effect.
