With the growth of population and the decrease of farmlands, food resources, esp. protein resources became more and more limited, so it was of great importance to explore protein resources largely. The synthetic exploration and utilization of slaughtering by-products was one of effective channels to solve this problem. Porcine blood, one cheap by-product full of protein that could be obtained, had about 18% protein,8 amino acid for adults and 18 amino acid including Histidine for babies, so it was one animal protein resource worth of exploration and utilization. At present, the synthetic utilization of porcine blood was not enough, except few of which was used for food or simply produced as feed, most of which was released to the environment causing only pollution. To explore a simple, convenient to practice, highly efficient and low-cost proteolysis of porcine blood was still one key to further exploration of porcine blood. The research used porcine blood as raw material, adopted hydrolysis of blood by highly-active Trypsin to focus on characters of proteolysis and proteolysis product, its evaluation of toxicity, as well as its functions like antioxidation, adjustment of blood fat, alleviation of tiredness. The main results of this research were as follows:1. Firstly proposing the adoption of optimal method to have proteolysis of porcine blood by fresh Trypsin to ensure optimal conditions of reaction:pH was 8.2±0.1, substrate containment was 6.5%, temperature was 39℃, [E]/[S] was 3%, time was 240 min; obtaining equation of RSM about DH and the above four factors: Y=-411.76+86.97A+2.68B+2.63C+8.74D+0.36AB+0.05AC-0.14AD+0.01BC-5.2381 0E-003BD-0.02CD-5.50A2-0.47B2-0.04C2-1.11D2The repeating experiments proved DH was 21.26%, PCL was 4.7, PR was 91.12% under the condition of proteolysis.2. By analyzing nutrient of PBH, it had high percentage of protein. Crude lipid, carbohodrate, mineral were of low percentage. PBH was basically similar to porcine blood on amino acid pattern, in which the essential amino acid were 40.27%, esp. Lysine about 7.36%. 3. By analyzing molecular weight of PBH, it was the combination of oligopeptide and amino acid below 1000Da, in which the percentage of molecular weight within 912-372Da was 71.28%.4 By analyzing the functions of PBH like solubility, emulsive capacity and the foaming capacity under different conditions, environmental factors had great impact on functions of PBH, so the adoption of PBH as functional nutrient to food and feed should take environmental factors into consideration, because only with proper condition can it produce the best effect.5. Kun Ming mice and Wister rats were used to explore the toxicological assessment on safety of food. The result proved PBH was safe and adopted to food and feed.6. By antioxidation experiment of PBH in vitro and in vivo to research on antioxidation activity, the result showed that the PBH had relatively strong scavenging power for free radical and possessed reducing power in vitro experiment; that PBH had protective function for the liver’s damage caused by Bromobenzene in vivo experiment.7. Wister rats were given high-fat feed and meanwhile administered with PBH to observe the influence towards blood lipids. The result showed that PBH had the auxiliary function to regulate blood lipids. It was firstly proposed that molecular mechanism about regulating blood lipids might improve the expression of LPLmRNA in order to strengthen activity of LPL in liver.8. Kun Ming mice were given different dosage PBH to study the alleviating fatigue effect. It was firstly found that PBH had apparent alleviating fatigue effects.
