At present, the number of patients on the waiting list of organ transplantation is far more than the number of organ for grafting. Moreover, the Utility of the available is not satisfied even the organ is preserved at low temperature. Of course, the graft cannot be biologically modified under this condition.Simulation of physiology condition for organ preservation should be the breakthrough in this field. In this study, auto-control techniques are used for the construction of the apparatus for organ preservation. Temperature, humidity, oxygen concentration, carbon dioxide concentration are auto-controled. Meanwhile, the idea of gene transferring for modification of graft has also been introduced into our apparatus. Based on this preliminary apparatus, we tested the relative criteria of this machine. The results thus obtained are as follows:1,We completed the platform for organ preservation study, this platform can control the temperature, humidity, oxygen concentration, carbon dioxide concentration. The range of temperature control is from - 20℃ to +60 ℃, the range of carbon dioxide concentration control is from 0% to 20%;the range of oxygen concentration control is from 2% to 98%;the range of humidity control is from the humidity at room temperature to 98%。2,The platform for organ preservation can be used for cell,tissue and bacteria culture; And it can be also used for small animal experiment at various temperature and humidity. 3,The platform for organ preservation was module designed. A different experiment could support by the correspongding module design. 4,The preservation apparatus for grfts was fixed successfully. The machine can be used for perfusion preservation in vitro. The temperature and humidity of storage chamber can be intellectually controlled, as well as the temperature and flow speed of perfusion.5,The application of the platform for organ preservation and the preservation apparatus for organ indicated that ① Cells such as 293 and PK15 cell lines could be successfully cultred in this machine.When temperature was going down as designed,the morphological changes of cells could be found;② The transfection efficency of Ad- PDL-1-EGFP was parralled to the temperature;③ the outcome of kidney perfusion at 8℃ was better than that at 20℃,as demonstrated by the histological observation,water content and apoptosis exam,etc;④ This apparatus could be easily used for graft perfusion.During the perfusion, the temperature could be either raised or lowered quickly, which made the higher gene transferring efficiency in graft.In summary, we for the first time made the apparatus for graft preservation and perfusion. And this platform could be used for gene transferring and increasing the quality of graft storage.
