Micro-channel Electrophoresis Chip (MCE Chip) system is a newly developed research field. The micro-channel network on the chip is manufactured by the technology of Micro Opto Electro Mechanical System (MOEMS) on the substrate of glass, fused silica, PDMS etc. Sample separation is performed in the micro-channel and detected by Opticcal or Chemical methods. Based on Capillary Electrophoresis (CE), MCE chip is a multidisciplinary technology, which has roots in Micro Opto Electro Mechanical System (MOEMS), biochemistry and analytical chemistry. It became a new platform in the field of analytical chemistry, especially in the life science, drug discovery and medicine.In this dissertation, the principle of MCE chip and development of MCE chip system are discussed. Then the method and the application of MCE chip system are studied. A new MCE chip system is also developed. The experiment results showed that using this system has successfully separated DNA segments.The topics of this dissertation are presented below:1. The capillary electrophoresis principle and the recent developments of MCE chip are reviewed. On the base of it, a novel design of MCE chip system is presented.2. Due to the inert to most chemicals, fused silica is chosen as the substrate and fabricated the chip using photolithography, wet etching and bonging technique. The inner side of micro-channel is smooth and straight. It is good for the experiments. Then the inner side of the micro-channel has been coated with linear polyacrylamide to decrease adsorption and the effecting of EOF.3. Two kinds of method of preparing for sieving matrixes are introduced. Two kinds of matrix injection method are designed and performed.4. The method of preparing and modifying analysis sample are introduced.5. The electrophoretic method of MCE chip is designed and performed. The electric field distribution at the cross is simulated and analyzing using the soft of ANSYS5.7. Then the electrophoretic method has been optimized. The electrophoresis and the volume of injection have been significantly controlled using this method.6. The migration characteristic of sample by the effect of electric field has been studied. The factors that cause the sample band broadening are discussed. Controlling the concentration of the buffer performs the sample stacking at the injecting cross. Then the effect of turning channel has been studied.7. This detection system has been used to DNA segments separation experiments; the results showed that it has met the need for DNA separation.The design and fabrication of LIF detecting system has been introduced in brief.The study of this dissertation showed that MCE chip has some advantages such as high speed and sensitivity. Because there are so much factors effecting the resolution and so complex of migration characteristic that it is hard to carry out DNA sequencing. It has to further optimize the electrophoretic conditions.Great progress in several related techniques, MCE chip has a broad application in the future.
