| Objective: To prepare the rat model of multiple organ dysfunction syndrome by intestinal ischemia-reperfusion, observe the level of serum endotoxin, the histomorphology of ileum and the express of P53 in gut immune barrier, so as to explore the mechanism of the multi-effects of Xuebijing injection(XBJ) on the immunologic barrier of intestinal ischemia-reperfusion rat.Methods: (1) Wistar rats were divided into 4 groups. including the control group, model group1, model group2, model group3. The superior mesenteric arteries were occluded for 30minutes, 45minutes and 60 minutes respectively. To establish an animal model of multiple organ dysfunciton syndrome (MODS) induced by ischemia-reperfusion of intestine, The serum endotoxin and some biochemical indicators were detected, The pathologic changes in different organs were examed scales, The death rate on 72h was also observed. and the occluded time was decided. (2) Wistar rats were divided into 4 groups randomly, including the control group, the model group, the ANT group, theXBJ group. 24h, 48h and 72h after the operation, the level of serum endotoxin(TNF-a,IL-1 and IL-6) were determined by ELISA method; the histomorphology of ileum were observed by HE method, includeing the thickness of intestinal mucosal, the height of intestinal villus and the intestinal mucosal injury index. (3) Wistar rats were divided into two groups randomly. including the control model group and the XBJ group. 24h, 48h and 72h after the operation, The express of P53 gene of CD4~+, CD3~+, CD8~+ T lymphocytes of ileal mucosal and laminapropria of ileal mucosal tissue were observed by immuohistochemical method, further more, the difference between the control model group and the XBJ group were compared.Results: (1) The level of serum endotoxin and UREA, CREA, ALT in model groupMG2 and MG3) were increased obviously(P<0.01). Many important internal organs appeared obvious pathological lesions. The results accorded with the criteria of MODS. The death rate on 72h of three model groups were 10%, 20% and 30%. The result was the best when the superior mesentery artery was occluded for 45 minutes. (2) After the operation, the level of serum TNF-a, IL-1 and IL-6 of the model group were higher than those of the control group(P<0.01). the level of serum TNF-a of the model group on the 72h were higher than those of the ANT group(P<0.01). the level of serum IL-1 of the model group on the 48h and 72h were higher than those of the ANT group(P<0.01). the level of serum IL-6 of the model group on the 24h, 48h and 72h were higher than those of the ANT group(P<0.01). the level of serum TNF-a, IL-1 and IL-6 of the model group and the ANT group were higher than those of the XBJ group(P<0.01). The thickness of intestinal mucosal, the height of intestinal villus in the model group were lower than the control group, the Chiu score of the model group were more than the control group(P<0.01). All the indicators of the ANT group were better than those of the model group(P<0.01). All the indicators of the XBJ group were better than those of the the ANT group(P<0.01). (3) The express of P53 of CD4~+, CD3~+ T lymphocytes and IgA plasmocytes in control group is higher compared with it in experiment group on the 48h and 72h (P<0.05,P<0.01); The express of P53 of CD8~+ T lymphocytes in control group has no difference compared with it in experiment group(P>0.05). In the control group, the express of P53 of CD4~+ T lymphocytes on the 72h is higher compared with it on the 24h and 48h(P<0.01); The express of P53 of CD3~+ T lymphocytes on the 72h is higher compared with it on the 24h(P<0.01); The express of P53 of IgA plasmocytes on the 72h is higher compared with it on the 24h and 48h(P<0.05). In experiment group, The express of P53 of CD3~+ T lymphocytes on the 72h is lower compared with it on the 24h(P<0.05).Conclusion: The animal model of MODS was established successfully by occluding the superior mesenteric arteries of rats for 45 minutes. Xuebijing injection could significantly decrease the level of serum TNF-a, IL-l and IL-6 in intestinal ischemia-reperfusion rat; it could relieve the lesion of intestinal mucosa from intestinal ischemia-reperfusion significantly; it can inhabit the express of P53 in gut immune barrier rises during the gut barrier injury; it may effect immune barrier of intestinal wall by this means. |
