Abstract/Summary:
Objective:The rabbit animal model of acute pulmonary thromboembolism ischemia-reperfusion successfully established by implanting2.7F coronary artery balloon catheter embolization with pulmonary artery. To observe the expression changes of rabbit model of TNF-a and mRNA in blood of acute pulmonary embolism ischemical reperfusion, the morphological changes of lung tissue combined with Xuebijing injection, clear protective effect on acute lung ischemia-reperfusion injury, to explore the mechanism of Xuebijing injection in the protection of acute lung ischemia-reperfusion injury, for the prevention and treatment of TCM lung ischemia-reperfusion injury theoryMethods:Healthy adult rabbits30, half male and half female, were randomly divided into normal control group (group A), ischemia-reperfusion group (group B), Xuebijing premedication day group (group C1), Xuebijing preoperative medication three days group (group C2) and Xuebijing operation six days before continuing to use the drug group (group C3) were divided into5groups,6rats in each group. A group of open pulmonary artery during the operation in2.7F coronary artery balloon catheter placed in the left, not embolization of the left pulmonary artery; B group in the preoperative day6after intravenous infusion of normal saline4mL/kg, pulmonary artery with balloon catheter embolization of the left lower intraoperative; C1, C2, C3group respectively before operation1days,3days,6days after the ear vein infusion of Xuebijing injection4mL/kg+normal saline, once a day, operation method with the B group. The rabbits respiratory rate, heart rate, the experimental process of30min/after reperfusion, choose30min observation results were compared between the two groups; each group before operation,2hours after embolization, after ischemia, reperfusion after2hours,4hours after reperfusion5times via ear vein serum, the level of TNF-α determination of ELISA;30min after reperfusion in collecting blood samples of RT-PCR detection of TNF-α mRNA expression; reperfusion for4hours after the death of animal, morphological changes were observed by light microscopy of lung tissue in FIE stainin.Results:1.30healthy adult rabbits, lung ischemia-reperfusion models24,18, and6rats died, after modeling pulmonary angiography showed satisfactory results, the success rate of the model was75%.2. Changes of respiratory frequency, heart rate:A group of rabbits in the experimental process of respiration and heart rate without exception. B, C1, C2, C3group, A group, breathing heart rate significantly faster, reached the peak at30minutes after reperfusion, then decreased slowly. Choice of reperfusion after30min observation results were compared between the two groups, B group of breathing, heart rate fast, group A was the slowest, between C1, C2, C3group, heart rate, respiration ranged from A to B group, C3group was the slowest in three groups. C1, C2group and B group were not significantly different between the two groups (P>0.05), no statistical significance between the two groups (P>0.05); C3group and A group were not significantly different (P>0.05), there were significant differences as compared with B, C1, C2group (P<0.05), respiratory rate, heart rate changes of C3group experimental process the minimum, the most close to the normal level.3. The serum TNF-a levels:changes at each time point in group A TNF-α level is not obvious, and there was no significant difference (P>0.05). B, C1, C2, C3group, TNF-a levels in pulmonary embolism after2hours than bolt pre-match increased, the difference was statistically significant (P<0.05), reperfusion after30minutes to reach the peak, then decreased. Selection of reperfusion after30minTNF-a levels were compared between the two groups, B group was the highest, the lowest C1, C2between group A, group C3between A, B group, C3group is the lowest among the three groups. C1, C2group and B group than there was significant difference between the two groups (P<0.05), no significant differences between the two groups (P>0.05); group C3and A, B, C1, C2groups were significantly different (P<0.05), suggesting that TNF-a levels and lung ischemia-reperfusion injury, Xuebijing injection can decrease serum levels of TNF-α, TNF-α level decreased with the duration of a certain corre.4. The expression of TNF-α4mRNA in blood:30min after reperfusion of blood TNF-α mRNA expression. RT-PCR showed expression of TNF-a mRNA in each group, the results of semi-quantitative data display, lowest in group A, group B was the lowest, between C1, C2, C3group between A, B group, C3group is the lowest among the three groups. C1, C2group and B group than have obvious difference (P<0.05) group, no significant differences between the two groups (P>0.05); there is no obvious difference between C3group and A group (P>0.05), compared with B, C1, C2groups were significantly different (P<0.05), suggesting that TNF-a mRNA expression is associated with lung ischemia-reperfusion injury, Xuebijing injection can reduce the expression of TNF-α mRNA. Blood and medication time has a certain correla.5. The pathological changes of lung tissue:the A group had no obvious lesions, B group showed diffuse pathological changes, marked hyperemia, edema, alveolar wall thickening, inflammatory cell infiltration, alveolar exudate, a large number of red blood cells and other components; lung pathological changes were not significantly different between C1group and B group, C2group, pulmonary edema and the exudation of light in the B group, C3group, pulmonary edema and inflammatory exudate significantly lighter than the C2group, C3group the degree of pathological changes in the light.Conclusion:The use of2.7F catheter in coronary artery to establish acute pulmonary thromboembolism ischemia-reperfusion rabbit animal model, strong controllability, animal mortality rate is low, can objectively reflect the characteristics of pulmonary ischemia reperfusion injury. Xuebijing injection can reduce the expression of TNF-α concentrations and TNF-α mRNA, reduce the lung tissue edema and intra-alveolar exudates, on lung reperfusion injur.
