The Effect Of Huoxue Qingxia Fa On The Gut Immunologic Barrier Of Intestinal Ischemia-Reperfusion Rat

Abstract/Summary:
Objective To prepare the rat model of multiple organ dysfunction syndrome by intestinal ischemia-reperfusion, observe the multi-effects of Huoxue Qingxia Fa on the immunologic barrier of intestinal ischemia-reperfusion rat and verify the enhance effect of Huoxue Huayu Fa to Tongli Gongxia Fa. Methods The MODS animal model was established by intestinal ischemia-reperfusion. After the treatment of Huoxue Qingxia Fa, the intestinal histomorphology was observed, the level of serum endotoxin and cytokines were determined. The level of slgA in intestinal contents and the quantity of gut associated lymphoid tissue were investigated. Results The level of serum endotoxin and cytokines of HXQX group were much lower than those of the model control group. The level of slgA in intestinal contents, the number of CD3+ and CD4+ lymphocyte and the ratio of CD4+/CD8+ of intestinal mucosa were higher in HXQX group than model control group. Conclusion Huo xue Qingxia Fa could protect the gut immunologic barrier of intestinal ischemia-reperfusion rat significantly. Huoxue Huayu Fa could enhance the effect of Tongli Gongxia Fa when the intestinal ischemia-reperfusion occurred.Objective To investigate the protective effect of Huoxue Qingxia Fa on the apoptosis of intestinal cell and the intestinal barrier in gut ischemia-reperfusion rat. Methods The rats were divided into 6 groups randomly, including the normal control group, model control group, ANT group, JWXCQ group, FDS group and HXQX group. 6h, 24h and 72h after the operation, the apoptosis index of intestinal mucosal epithelial, lamina propria and folliculus lymphaticus were investigated. The expression of Apaf-1, Caspase-3, Bcl-2 and COX-2 mRNA in intestinal mucosal cells were observed. Results The apoptosis indexes of intestinal mucosal epithelial, lamina propria , folliculus lymphaticus of the model control group were higher than those of the normal control group. The expression of Apaf-1, Caspase-3 and COX-2 mRNA were higher in the model control group than the normal control group, while the expression of Bcl-2 was lower. All the indicators of the HXQX group were better than those of the model group. Conclusion Huoxue Qingxia Fa could significantly protect the gut barrier in intestinal ischemia-reperfusion rat by lowering the AI of epithelial cell and lymphoid cell, decreasing the expression of Apaf-1, Caspase-3 and COX-2 mRNA, and increasing the expression of Bcl-2.
