The Theoretical And Experimental Research On The Effect Of Delaying Senescence By Jingui Shenqi Wan

Delaying senescence is the everlasting dream for everyone.In the modern society,the increasing average life expectancy and the senescence of population make people more aware of senescence and longevity.The scientific study of life is focusing on delaying senescence,which can not only prolong the average life expectancy,but also delay the senescence process of the middle-aged and elderly and improve their qualities of life.The research on regimen and delaying senescence has a long history in China,which can date back to the Pre-Qin Period.The development of TCM in the past thousands of years led to some unique cognitions and valuable theories on senescence,developing various distinctive approaches for delaying senescence and regimen as well as many well-known traditional formulae that has long been widely used by the doctors with undoubted curative effects.The enforcement of the research on their delaying senescence and enlarging their clinical application is of great significance as to the people's needs for delaying senescence.The research on delaying senescence is bound to draw increasing attention from medical circle because of the clinical superiority and potential of TCM.This research project aims at exploring the mechanism of "Jingui Shenqi Wan" in delaying senescence.The theoretical research falls into three main parts:First,based on the comprehensive analysis on the main theories of senescence in TCM as well as the findings of modern theoretical and clin cal research,we put forward the theory of kidney deficiency and phlegm-stasis senescence as the central theory of senescence in TCM.According to this theory,kidney deficiency is the basic cause of senescence,while phlegm rheum and blood stasis are the leading pathological symptoms caused by kidney deficiency in the process of senescence,which will in return aggravate kidney deficiency and quicken the process of senescence,kidney deficiency and phlegm stasis act together and eventually cause the decline or even failure of the Viscerae and Bowels function.Second,this research makes an analysis on the relations between the theory of kidney deficiency and phlegm-stasis senescence and other senescence theories such as the five viscera deficiency senescence,spleen deficiency senescence,Ying-yang disharmony senescence and the essential qi and spirit deficiency senescence.The research finds that the theory of kidney deficiency and phlegm-stasis senescence can cover all the other theories.The analysis on the modern findings of senescence theory in TCM also shows that this theory,living up to the congition of modern life science on senescence,is connected more closely and extensivly to the modern theory of senescence.Third,this research also provides a fruitful discussion on the TCM mechanism of the effects of delaying senescence by "Jingui Shenqi Wan".As the classic formula of supplementing the kidney and prolonging life,this formula has been widely used by doctors in regimen,delaying senescence and the treatment for the old patients.The current documents report nothing about the research on the TCM mechanism of this formula on delaying senescence.This research explores the significance of Jingui Shenqi Wan compatibility,some original text from Synopsis of Prescriptions of the Golden Chamber,the original formula evolution and modern clinical application,and put forward that the main curative effects of Jingui Shenqi Wan lies in supplementing kidney qi and transforming stasis and phlegm.Its curative effects is in accordance with the pathological mechanism of senescence,kidney deficiency and phlegm stasis,which is the key of the markable effect of delaying senescence.Also,based on the analysis of the ingredents and the compatlity significance,this research advocates that the significance of compatility is not "pursuing yang in ying"The experimental research summarizes the previous formula(methods) of supplementing kidney and the data on "Jingui Shenqi Wan" in delaying senescence.Based on this study,this research,resorting to the method of serum pharmacology of TCM, studies the effects of Jingui Shenqi Wan on the human diploid fibroblast shape of people at different ages and on the membrane potential of human diploid fibroblast mitochondrion and the nucleolus DNA of the human diploid fibroblast.The findings are as follows:(1.) the change of the shape of cell surface shows that "Jingui Shenqi Wan" serum can impove the living condition of human diploid fibroblast and slow down senescence.(2.) the analysis through Flow Cytometry indicates that this formula can change the electrode of human diploid fibroblast mitochondrion and decrease the death rate of cells;(3.) Observing the chromogenic reaction through microscope and judging the effects of the medicine on the death of cells suggest that this formula can decrease the rupture of human diploid fibroblast DNA so as to prevent the death of cells.(4.) This formula can prevent the death of cells by enhancing the expression of Bcl-2 and adding more Bcl-2 protein to the cells to prevent mitochondrion from giving off cell pigments.This research deepens the previous researches on the effects of Jingui Shenqi Wan on delaying senescence and explores the effects of delaying senescence in terms of molecule mechanism.The findings show that Jingui Shenqi Wan can decrease the death of cells,which is likely to be the vital mechanism of its effects of delaying senescence.This research promotes the recoginiton of the effects of delaying senescence by Jingui Shenqi regarding modern scientific mechanism.This research also clarifies the theoretical bases of the effects of Jingui Shenqi Wan on delaying senescence,further defining the modern scientific mechanism of this curative effect and providing the basis for expanding its clinical application.Also,this research enriches the cognition of the mechanism of delaying senescence by supplementing kidney and tranforming stasis and phlegm,which is helpful for the further understanding of kidney deficiency senescence and kidney essence.
