| Objective:Ob serving the effects of Bushen-Jianpi formula on nuclear factor kappa-B(NF-κB),Sirtuin 1(SIRT1),Sirtuin 6(SIRT6)of aging rats with kidney deficiency syndrome and spleen deficiency syndrome,SIRT6)expression and the level of inflammatory factors,to explore the potential mechanism of bushenj ianpi formula to delay immune aging.Methods:56 healthy male rats were randomly divided into 8 groups,7 in each group.D-gal actose was used to replicate the aging model.On the basis of the aging model,hydrocortisone and rhubarb were given ingavage to establish the aging model of kidney deficiency syndrome and spleen deficiency syndrome;the other groups were given the same amount of normal saline.Serum Superoxide di smutase(SOD)content was detected by xanthine oxidase method,determination of serum Adrenocorti cotropic hormone(ACTH),Amylase(AMS),Interleukin-1beta(IL-1β),Interleukin-6(IL-6),Tumor necrosis factor-alpha(TNF-α)content by enzyme-linked immunoassay,HE staining was used to observe the pathological morphology of kidney and spleen,the expressions of NF-κB,SIRT1 and SIRT6 in kidney and spleen were observed by Wersten-blot and immunohistochemistry.Results:Compared with blank group,SOD in senescence group,kidney deficiency senescence group and spleen deficiency senescence group decreased significantly,which proved that the senescence model was successfully constructed.Compared with the aging group,ACTH in the kidney deficiency aging group was significantly decreased,which proved that the aging model of kidney deficiency syndrome was successfully constructed;AMS in the spleen deficiency aging group was significantly decreased,which proved that the aging model of spleen deficiency syndrome was successfully constructed.Compared with kidney deficiency and senility group,ACTH,IL-1β,TNF-α,IL-6,NF-κB expression,SIRT1 and SIRT6 expression were increased significantly in Jinqui shenqi Wan group and kidney deficiency Bushenj ianpi prescription group.Compared with spleen deficiency aging group,AMS,IL-1β,TNF-α,IL-6,NF-κB expression and SIRT1 and SIRT6 expression were significantly increased in Sijunzi Decoction group and spleen deficiency Busshen Jianpi prescription group.Conclusion:Bushen Jianpi Decoction can delay immune aging by inhibiting the immune inflammatory response mediated by NF-κB/SIRT1/SIRT6 signaling pathway and maintaining the balance of inflammatory factors IL-1β,TNF-α and IL-6. |
